摘 要:目的:研究卡介苗对阿尔茨海默病(AD)模型小鼠行为及炎症因子水平的影响。方法:将32只成年C57BL/6小鼠随机分成四组,分别为对照组、AD组、卡介苗低剂量组(104CFU)和高剂量组(105CFU)。除对照组外,其他三组小鼠均以连续腹腔注射D-半乳糖建立AD模型,其中两组施以不同剂量的卡介苗。各组小鼠通过旷场和水迷宫检测其活动度和学习记忆功能;免疫荧光检测脑内Aβ斑块; ELISA检测血清中炎症因子TNF-α和IFN-γ的含量。结果:与对照组相比,AD小鼠活动度下降,学习记忆能力退化,脑内可检测到Aβ沉积,血清中TNF-α、IFN-γ含量上升,而经过卡介苗处理的AD小鼠,不论剂量高低,其行为学均得到改善(P<0.05),脑内Aβ斑块减少(P<0.05),血清中TNF-α和IFN-γ含量降低(P<0.05)。结论:低剂量和高剂量的卡介苗均能改善AD小鼠的行为,降低小鼠脑内Aβ沉积,控制血清中的炎症因子。Objective :To obsetne the impact of Bacillus Calmette-Guerin (BCG) on the behavior and inflaramatory factors of model mice with Alzheimer disease(AD). Methods:Thirty-two adult C57BL/6 mice were equally divided into control group, AD group, low BCG (104CFL) and high BCG ( 105 CFL ) treatment groups. AD mouse models were induced by continuous intraperitoneal injection of D-galaetose in three groups except the controls, and different dosage of BCG was applied to two groups of model mice. Then mice in each group were subjected to open-field test and Morris water maze test to observe the activities and the learning and memory abilities, Immunofluoreseenee stain was used to detect amyloid-beta(Aβ)-associated cerebral plaques, and ELISA was performed to measure levels of inflammatory factors TNF-α and IFN-γ Results : Compared with the controls, AD mice had decreased activi- ty and ability of learning and memory,and Aβ deposition could be observed in the brain with increased serum levels of TNF-α and IFN-γ. l lowever, the mice had improved behaviors as well as decreased deposition of Aβ and TNF-α and IFN-γ levels following treatment with BCG either in low or high dosage (all P〈 0.05). Conclusion:Either low or high dosage of BCG can improve the behaviors in AD mouse, reduce the Aβ-assoeiated plaques and regulate the serum inflammatory factors.
