摘  要:目的:研究N-myc下游调节基因2(NDRG2)对卵巢癌迁移和侵袭能力的影响。方法:首先应用qRT-PCR实验检测NDRG2在正常卵巢组织和卵巢癌组织中的表达水平。再应用划痕实验和transwell小室实验检测NDRG2对卵巢癌细胞侵袭和迁移能力的影响。最后检测NDRG2对SKOV3细胞中基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶-9(MMP-9)表达水平的影响。结果:在卵巢癌组织中NDRG2的表达水平明显低于正常卵巢组织。NDRG2的表达水平可以影响卵巢癌细胞的侵袭和迁移能力,NDRG2减少MMP-2和MMP-9的表达水平。结论:NDRG2在卵巢癌中作为一个抑癌基因可以影响癌症的侵袭和转移。Objective: N-myc downstream regulated gene 2( NDRG2) is a candidate gene for tumor suppression.The expression of NDRG2 is down-regulated in several tumors.The aim of this study was to explore the effect of NDRG2 on invasion,migration,and enzymatic activity of matrix metalloproteinase-2( MMP-2) and-9( MMP-9) in human ovarian cancer SKOV3 cells.Methods: We investigated the expression of NDRG2 in ovarian cancer tissues and normal ovarian tissues by qRT-PCR.The effects of NDRG2 expression on cell invasion and migration were evaluated using transwell filter migration assay and wound healing assay. The expression of MMP-2 and MMP-9 was measured by western blot assay.Results: The level of NDRG2 in ovarian cancer was lower than normal ovarian tissues.The findings of wound healing and transwell assay showed that NDRG2 reduced migration and invasion of SKOV3 cells compared to control cells.Western blot analyses revealed that NDRG2 decreased the expression of MMP-2 and MMP-9.Conclusions: These findings suggest that NDRG2 may be a new anti-invasion factor in ovarian cancer through inhibiting MMPs activities.
