摘 要:目的 探讨在哮喘动物模型中microRNA-155(miR-155)的表达变化及其对调节性T细胞(regulatory T cell,Treg)与Th17细胞比例的影响.方法 应用卵清蛋白致敏C57BL/6小鼠的方法建立小鼠哮喘动物模型组并设置对照组,分离、获取肺周围淋巴结以及脾组织的混合T淋巴细胞,进行体外淋巴细胞培养,并同时建立antago-miR-155拮抗封闭组,应用流式细胞分析检测各组T淋巴细胞亚型的变化.结果 成功建立了C57BL/6哮喘小鼠动物模型,分离培养淋巴细胞后进行流式细胞分析检测,miR-155封闭组与对照组相比,在封闭miR-155之后,动物模型的Treg细胞所占百分比(3.93±0.27)%显著增高而Th17细胞百分比(0.77±0.01)%显著降低(P〈0.05).结论 哮喘动物模型中miR-155可调节免疫系统的Treg和Th17的细胞平衡,为研究哮喘疾病治疗的新靶点奠定基础. Objective To investigate the effect and immunomodulation mechanism of miR-155 on reg-ulatory T cell(Treg)and Th17 ratio in asthma animal model. Methods C57BL/ 6 mice were sensitized and challenged with OVA to establish asthma animal model. Lymphocytes were isolated at the peak period of the dis-ease. Lymphocytes were cultured with or without antago-miR-155. The percentage of Th cells were evaluated by flow cytometry. Results After establishment of asthma animal model,antagomiR-155 group compared with con-trol group,the proportion of Treg (3. 93 ± 0. 27)% increased while Th17(0. 77 ± 0. 01)% decreased. Conclu-sions miR-155 strongly reduces Treg generation and promote Th17 cell generation which play an important role in the pathogenesis of allergic asthma,ensuring the basic foundation of new targets for the treatment of asthma.
