摘  要:目的研究褪黑素对大鼠肺缺血再灌注损伤的保护作用及其可能机制。方法选取成年雄性大鼠36只,建立大鼠肺缺血再灌注损伤模型,依次分为假手术组,缺血再灌注组以及褪黑素预处理组,分别取各组肺组织检测超氧化物歧化酶,丙二醛,再通过检测肺组织湿干重比以及支气管肺泡灌洗液中总蛋白含量用以衡量褪黑素预处理对大鼠肺缺血再灌注损伤的保护作用。结果缺血再灌注组丙二醛明显高于假手术组,并且其肺组织湿干重比以及支气管肺泡灌洗液中总蛋白浓度明显上升,抗氧化指标超氧化物歧化酶表达呈降低趋势(P<0.05),但褪黑素预处理能够明显缓解由缺血再灌注损伤诱发的肺组织病理变化,丙二醛、肺组织湿干重比以及支气管肺泡灌洗液中总蛋白的浓度呈明显下降趋势且,超氧化物歧化酶水平升高且数值具有统计学意义(P<0.05)。结论褪黑素能够通过直接或间接清除自由基而对肺组织发挥保护作用。Objective Study on the protective effect of melatonin on rat lung ischemia-reperfusion injury and its possible mechanism. Methods 36 adult male rats were selected to establish rat model of lung ischemia-reperfusion injury, It was divided into sham operation group, ischemia reperfusion group and melatonin preconditioning group. Detection of superoxide dismutase and malondialdehyde in each group of lung tissues, and the protective effect of melatonin pretreatment was measured by detecting the ratio of wet dry weight and the total protein concentration in bronchoalveolar lavage fluid. Results The malondialdehyde in the ischemia-reperfusion group was significantly higher than that in the sham operation group, and the ratio of wet dry weight and the total protein concentration in bronchoalveolar lavage fluid increased obviously, Furthermore the expression of superoxide dismutase is decreasing(P〈0.05). However, melatonin preconditioning could significantly alleviate the pathological changes of lung tissue. The malondialdehyde, wet dry weight ratio and the concentration of total protein in bronchoalveolar lavage fluid remarkable decreased,the superoxide dismutase(SOD) level is higher and the value is statistically significant(P0.05). Conclusion Melatonin can protect lung tissue by direct or indirect scavenging free radicals.
