摘　　要：目的探讨异黏蛋白(MTDH)在子宫内膜腺癌组织中的表达及其临床意义。方法采用免疫印迹(Western blot)与反转录-聚合酶链反应(RT-PCR)方法检测83例子宫内膜腺癌组织与50例正常子宫内膜组织中MTDH蛋白及mRNA的表达情况,分析MTDH的表达与子宫内膜腺癌患者临床病理特征及预后的关系。结果子宫内膜腺癌组织中的MTDH蛋白及mRNA的表达均明显高于正常子宫内膜组织,MTDH蛋白的过表达与腺癌组织的细胞分化程度、有无淋巴结转移、肌层浸润深度及FIGO分期有关,而与年龄、雌激素受体(ER)、孕激素受体(PR)、血CA125无明显相关性。Kaplan-Meier生存结果分析提示子宫内膜腺癌组织中MTDH蛋白过表达总生存期明显差于低表达组,多因素回归分析发现分化程度、淋巴结转移、FIGO分期、肌层浸润深度及MTDH表达水平这5项因素与子宫内膜腺癌患者的预后密切相关。结论 MTDH的过表达可能与子宫内膜癌增殖、侵袭、局部淋巴结转移和分期有密切关系,提示MTDH可能是根治性术后子宫内膜癌患者预后指标,可评价子宫内膜癌恶性生物学行为,为子宫内膜腺癌的治疗及预后判断提供参考。Objective （MTDH） in endometrial To investigate the expression and clinical significance of metadherin adenocarcinoma. Methods The expression of MTDH protein and gene mRNA were detected by Western blot and RT-PCR in 83 cases of endometrial adenocarcinoma and 50 cases of normal endometrial tissue.The relationships of MTDH expression to clinical path ological characteristics and prognosis were analyzed in patients with endometrial adenocarcinoma. Results Both mRNA and protein expressions of MTDH in endometrial adenocarcinoma tissue were significantly higher than those in normal endometrial tissue. The overexpression of MTDH protein was positively correlated with histopathological grade,lymph node metastasis, myometrial invasion depth and FIGO stage,but not with age,ER,PR and blood CA125.Kaplan Meier survival analysis demonstrated that the overall survival in MTDH overexpression group was significantlywere closely associated with the prognosis of endometrial adenocarcinoma.Conclusion The over expression of MTDH is closely related to the proliferation, invasion, lymph node metastasis and stage of endometrial adenocarcinoma, suggesting that MTDH may be a prognostic indicator in en dometrial adenocarcinoma patients after radical surgery,which can be used for the evaluation of malignant biological behavior and provides a new reference for treating endometrial adenocarcino ma and assessing prognosis
