摘  要:试验以矮小品系S2鸡为试验素材,采用q-PCR技术以及SPSS分析软件,研究焦点黏连(Focal adhesion)信号通路中调控:小凹蛋白-3(CAV-3)、血小板衍生生长因子受体α多肽(PDGFRA)、分泌型焦磷酸蛋白(SPP1)在生长期骨骼肌发育中的表达模式及在成肌细胞增殖和分化期的表达特征。结果显示:在肌肉组织中,CAV-3在胸肌和腿肌中的表达模式相同,从4周龄开始随着发育时间的增加,CAV-3表达量逐渐降低,在16周龄时显著低于其他发育时期(P<0.05);在0日龄、8周龄时PDGRFA在胸肌中相对表达量显著高于腿肌(P<0.05),但在16周龄时胸肌中PDGRFA相对表达量显著低于腿肌(P<0.05);SPP1在胸肌和腿肌中的表达模式存在显著差异,从4周龄开始,在胸肌中随着发育时间增加,SPP1相对表达量逐渐减少,在腿肌中随着发育时间的增加,SPP1相对表达量逐渐减少。在成肌细胞中,CAV-3相对表达量在增殖期100%时显著高于其他发育时期(P<0.05),并随分化时间的增加,CAV-3相对表达量逐渐减少;PDGFRA表达量在分化期的表达显著高于增殖期(P<0.05),并随诱导分化时间的延长表达量逐渐升高;SPP1相对表达量在增殖期100%时显著高于其他发育时期(P<0.05)。结果表明:CAV-3、PDGFRA和SPP1 mRNA表达量在鸡成肌细胞增殖分化期和鸡肌肉组织生长期分别呈现明显的差异性,3种基因可能分别以不同的方式参与调控胸肌、腿肌发育过程,为进一步研究该3种基因分别调控肌肉发育的分子机制提供了理论依据。In this study, expression characteristics of CAV-3, PDGEFA and SPP1 mRNA expression of focal adhesion signal pathwayin chickenmyoblasts and muscle tissues was detected using q-PCR and SPSS in the $2 strain. The result showed that CAV-3 mRNA expression in breast and leg muscle had the same pattern, CAV-3 mRNA expression peaked at 4 weeks of age and dropped by degrees, CAV-3 mRNA expression at 16 weeks of age was significantly lower than other developmental stages (P〈0.05). PDGRFA mRNA expression in breast muscle was significantly higher than leg muscle at 0 days and 8 weeks of age, but PDGRFA mRAN expression in breast muscle was significantly lower than leg muscle (P〈0.05). The SPP1 mRNA expression in breast and leg muscle had the significantly differential pattern. Starting at 4 weeks of age, SPP1 mRNA expression in breast muscle dropped with age, but SPP1 mRNA expression in leg muscle increased with age. In mynbiasts, CAV-3 mRNA expression in proliferation period of 100% was significantly higher than other developmental stages (P〈0.05), and CAV-3 mRNA expression graduallydropped with induced-differentiation time. PDGRFA mRNA expression in the differentiation period was significantly higher than that of proliferative period (P〈0.05), and PDGRFA mRNA expression was gradually increased with induced-differentiation time. SPP1 mRNA expression in proliferation period of 100% was significantly higher than other developmental stages (P〈0.05), but SPPI mRNA expression had no significant change with induced-differentiation time. Combined with CAV-3, PDGRFA and SPP1 mRNA expression in muscle tissue and myoblasts, these findings suggested that 3 related genes mRNA expressionof focal adhesion signal pathway in skeletal muscles presented significant breed and tissue difference, and they may be involved in the regulation of breast and leg muscles development in different ways. This study laid a foundation to elucidate the genetic mechanisms of muscle growth in chicken.
