青海地区藏族HIF-2α基因单核苷酸多态性与胃癌易感性

摘　　要：目的探讨青海地区藏族HIF-2α基因单核苷酸多态性与藏族胃癌易感的相关性。方法收集世居青海的健康藏族62例作对照组、胃癌患者45例作胃癌组。利用血液基因组DNA提取试剂盒提取并纯化被研究者外周血。采用PCR及测序法检测HIF-2α基因的rs13419896、rs7598371、rs6715787位点,并行相关性分析。结果 rs13419896SNP基因型分别为A/G、G/G、A/A,rs7598371SNP基因型分别为C/G、C/C、G/G,rs6715787SNP基因型分别为C/G、C/C、G/G。对照组检验基因型分布符合Hardy-Weinberg平衡规律(P>0.05)。其中rs13419896的A/G、G/G、A/A基因型频率在胃癌组中分别为28. 89%、28. 89%、42. 22%,在对照组中的频率为38. 71%、27.42%、33.87%。rs7598371的C/G、C/C、G/G基因型频率在胃癌组分别为26. 67%、35. 56%、37.78%,在对照组分别为41.94%、25.81%、32.26%。rs6715787的C/G、C/C、G/G基因型频率在胃癌组分别为35.56%、40.00%、24.44%,在对照组分别为40.32%,32.26%、27.42%。经χ~2检验示,胃癌组与对照组间无统计学差异(P>0.05)。结论青海地区藏族HIF-2α基因单核苷酸多态性与藏族胃癌易感性无关。Objective To study the association between the single nucleotide polymorphism of Tibetan HIF-2α gene and susceptibility to gastric cancer in Qinghai.Methods A total of 62 Tibetans（ peripheral blood）and 45 gas-tric cancer patients（peripheral blood）of Tibetans living in Qinghai Province were collected.DNA was extracted and purified using conventional phenol-chloroform-isoamyl alcohol method and DNA extraction kit.The rs13419896, rs7598371, and rs6715787 loci were used as the analysis sites for polypeptides.PCR method was used to analyze the polymorphism changes.Afterwards,the polymorphic products were randomly selected for sequencing to verify the ac- curacy of the analysis.Results According to the test results of the patient specimens ,the rs13419896 polymorphic lo- ci were A/G, G/G, and A/A, respectively. The rs7598571 polymorphic loci were mainly C/G, C/C, and G/G. rs6715787,respectively.The polymorphic sites are mainly C/G, C/C, and G/G.The genotype distribution was ac- cordance with Hardy-Weinberg equilibrium（P〉0.05）.The frequency of AJG, G/G, and A/A in rs13419896 was 28.89%, 28.89%, and 42.22%, respectively, in the gastric cancer group ; and 38.71%, 27.42%, and 33.87% in the control group.The frequencies of rs7598371C/G, C/C, and G/G in the gastric cancer group were 26.67%, 35.56%, and 37.78 %, respectively, and 41.94%, 25.81%, and 32.26% in the control group, respectively. The frequencies of C/G, C/C, and G/G in rs6715787 were 35.56%, 40.00%, and 24.44%, respectively, in the gastric cancer group ; and 40.32%, 32.26%, and 27.42% in the control group, respectively.There was no significant difference between the gastric cancer group and the control group by X2 test（P〉0.05）.Conclusion The risk of gastric cancer in Qinghai has no significant relationship with the single nucleotide polymorphism of HIF-2α gene.
