摘 要:目的通过离体实验的方法,观察-Gx加速度作用对家兔肺脏组织基质金属蛋白酶(matrix metalloproteinase,MMP) 9和基质金属蛋白酶抑制剂(tissue inhibitor of metalloproteinase,TIMP) 1表达水平的影响,探索-Gx致肺损伤的作用机制。方法选取1月龄的雄性新西兰家兔20只,随机分为实验组和对照组,每组10只。实验组动物采用水平加速度试验平台进行-Gx加速度作用,加速度作用峰值3. 6 G,持续时间为2 s,每天进行加速度作用20次,间隔5 min,连续30 d。对照组动物不进行加速度作用,其他处理同实验组。采用免疫组化方法检测各组动物肺组织MMP9和TIMP1蛋白含量变化情况。结果 (1)对照组动物肺组织每高倍镜视野MMP9阳性细胞数为(13. 7±7. 2),-Gx暴露组动物每高倍镜视野阳性细胞数为(28. 3±12. 5),与对照组相比,-Gx暴露组动物肺组织的MMP9表达水平显著增高(P<0. 05);(2)对照组动物肺组织每高倍镜视野TIMP1阳性细胞数为(17. 4±10. 2),-Gx暴露组动物每高倍镜视野阳性细胞数为(8. 6±5. 9),与对照组相比,-Gx暴露组动物肺组织的TIMP1表达水平显著降低(P<0. 05)。结论 -Gx水平加速度作用可导致家兔肺组织MMP9和TIMP1表达水平的改变,其可能参与了-Gx加速度作用致肺损伤的发生发展过程。Objective To investigate the effects of -Gx on matrix metalloproteinase (MMP) 9 and tissue inhibitor of metalloproteinase (TIMP) 1 expression in rabbit lung. Methods Twenty male rabbits at 1 month old were divided into control group and -Gx group. The -Gx group rabbits were exposured by -Gx acceleration with peak of 3.6 G and 2 s. Rabbits of -Gx group were exposured total 30 clays and 20 times per clay. The expression of MMP9 and TIMP1 were evalulated by immunohistoehemieal method. Results (1)The positive cell number of MMP9 in control group is (13.7±7.2), that in -Gx group is (28.3±12.5). The expression of MMP9 protein level in -Gx group rabbit is higher than that in control group (P〈0.05) ; (2)The positive cell number of TIMP 1 in control group is ( 17.4±10.2), while in -Gx group is (8.6±5.9). The expression level of TIMP1 protein in -Gx group is lower than that of in control group(P〈0.05). Conclusion The MMP9 and TIMP1 expression in rabbit lung eouht be changed by -Gx action, which eouht be involed with the mechanism of lung injuU induced by -Gx.
