摘 要:为探讨真菌诱导子与龙眼胚性愈伤组织中类黄酮、类胡萝卜素和单宁含量的相关性,以龙眼胚性愈伤组织细胞为材料,研究真菌诱导子种类、浓度及处理天数对龙眼愈伤组织细胞中类黄酮、类胡萝卜素和单宁含量的影响。结果表明:在固体培养基中加入100mg/L拟茎点霉菌诱导子培养35d时,最有利于类黄酮含量的积累,含量高达8.58 mg/g;拟茎点霉菌诱导子浓度为50 mg/L时培养35 d,最有利于类胡萝卜素含量的积累,其含量为36.83μg/g;胶孢镰刀菌诱导子浓度为50mg/L时的培养基中培养35d,最有利于单宁含量的积累,其含量为10.50 mg/g。液体悬浮培养条件下,胶孢镰刀菌诱导子浓度为400 mg/L的培养基中培养7 d,最有利于类黄酮含量的积累,其含量为6.29 mg/g;尖孢镰刀菌诱导子浓度为200 mg/L的培养基中培养7 d,最有利于类胡萝卜素含量的积累,其含量为49.21μg/g;胶孢镰刀菌诱导子浓度为400 mg/L的培养基中培养7 d,最有利于单宁含量的积累,其含量为13.15mg/g。本研究为将来龙眼胚性愈伤组织细胞工厂化生产类黄酮、类胡萝卜素和单宁等次生代谢物质奠定理论基础并提供技术指导。To study the correlation between fungi elicitors and flavonoids, carotenoids and tannins in the embryogenic calli of longan, the embryogenic calli were used as the materials to investigate the effects of different kinds of fungi elicitors for the contents of flavonoids, carotenoids and tannin in callus cells. The results showed that under the condition of solid culture, the flavonoid content was the highest(8.58 mg/g) under the treatment of 100 mg/L Phomopsis longanae Chi for 35 d, and the carotenoid content was the highest(36.83 μg/g) under 50 mg/L P. longanae Chi treatment. For tannin, however, the content was the highest(10.50 mg/g) under Fusarium subglutinans Wollenw. et Reinkin treatment for 35 d. Under liquid culture, the flavonoid content was the highest(6.29 mg/g) under the treatment of 400 mg/L F. subglutinans Wollenw. et Reinkin treatment for 7 d, the carotenoid content was the highest(49.21 μg/g) under the treatment of 200 mg/L F. Oxysporum for 7 d, and the tannin content was the highest(13.15 mg/g) under 400 mg/L F. subglutinans Wollenw. et Reinkin treatment for 7 d. The above results could provide a scientific basis and technical support for studying the metabolic mechanism of flavonoids, carotenoids and alkaloids.
