摘 要:目的:探讨红景天苷(SAL)对大鼠脑缺血再灌注损伤(IRI)的神经保护作用。方法:建立大鼠右侧大脑中动脉缺血再灌注模型。实验分为假手术组、IRI模型组、SAL预处理+IRI组(每日10 g/kg,溶于生理盐水,浓度为1. 5 g/ml,连续灌胃14 d),于再灌注24 h应用试剂盒检测大鼠缺血脑组织中丙二醛(MDA)、活性氧簇(ROS)及超氧化物岐化酶(SOD)的表达水平;应用2,3,5-氯化三苯基四氮唑(TTC)染色方法评定脑梗死体积,计算梗死体积百分比;用Western Blot和免疫荧光染色方法观察大鼠缺血侧脑组织神经元表达的变化。结果:与假手术组相比,IRI模型组MDA、ROS的表达水平明显升高,SOD的表达水平明显降低(P <0. 01),脑梗死体积百分比增大(P <0. 01),Neu N的表达降低(P <0. 01);与IRI模型组相比,红景天苷预处理+IRI组MDA、ROS的表达明显降低,SOD的表达明显升高(P <0. 05),脑梗死体积百分比明显减小(P <0. 01),Neu N的表达升高(P <0. 05)。结论:红景天苷可通过抑制脑缺血再灌注损伤大鼠的氧化应激来减少脑梗死体积而发挥神经保护作用。Objective: To investigate the neuroprotective effect of salidroside( SAL) on cerebral ischemia-reperfusion injury( IRI) in rats.Methods: A rat model of middle cerebral artery occlusion was established.The rats were divided into sham group,IRI group,SAL pretreatment + IRI group( salidroside dissolved in saline administered at a concentration of 1.5 g/ml by 10 g/kg/d for consecutive 14 days).The levels of malondialdehyde( MDA),reactive oxygen species( ROS) and superoxide dismutase( SOD) in ischemic brain tissue of rats at 24 hours after reperfusion were detected.The cerebral infarction volume was evaluated by using 2,3,5-triphenyltetrazolium chloride( TTC) staining method and the infarct volume percentage was calculated.Western Blot and immunofluorescence staining were used to observe the expression of Neu N in the ischemic brain tissues.Results: Compared with the sham group,the expression of MDA and ROS in IRI group increased significantly,while the expression level of SOD decreased significantly( P〈0.01),the percentage of infarction volume increased( P〈0.01) and the expression level of Neu N + cells decreased significantly( P〈0.01) at IRI group; In contrast,compared with IRI group,the expression of MDA andROS in SAL pretreatment + IRI group was significantly decreased,while the expression of SOD was significantly increased( P〈0.05),the percentage of infarction volume was significantly reduced( P〈0.01) and the expression level of Neu N + cells increased significantly( P〈0.05).Conclusion: Salidroside could inhibit oxidative stress and reduce cerebral infarction volume to exert neuroprotective effect on rats with cerebral reperfusion injury.
