摘 要:利用微纳加工技术加工出栅形孔氮化硅过滤膜,通过物理过滤实现了循环肿瘤细胞(CTCs)的高效捕获。为进一步研究CTCs,通过反向冲击将CTCs从过滤膜上释放,然而由于CTCs与过滤膜之间存在粘附力,导致释放效率较低。通过在过滤样品和反向冲击液中加入抗凝剂肝素,大大提高了CTCs的释放效率和过滤膜的可重复利用性,并且能保证释放出的CTCs的活性。利用该技术加工的过滤器件和释放方法,能够实现CTCs的高效捕获和释放,具有操作方便、低成本、高通量以及易于保证细胞活性等优点。A type of grating-shaped silicon nitride thin film filter was fabricated with micro-fabrication technology,the circulating tumor cells(CTCs)can be captured efficiently by physical filtration.In order to further study CTCs,it need to be released from the thin film filter by reverse flow.However,the release efficiency was too low,because of the adhesion between the CTCs and the filter surface.Through added of thinner heparin in the filter sample and reverse flow,the release efficiency of CTCs and the reusability of a filter are greatly improved,and the viability of CTCs can also be ensured.CTCs can be captured and released efficiently with the method proposed in this paper,it can be realized that efficient capture and release which has the advantages of easy operation,low cost,high throughput and easy to ensure cell viability.
