摘 要:目的:探讨骨髓间充质干细胞(BMSCs)移植联合中医电针对脑出血大鼠脑源性神经营养因子(BDNF)与神经生长因子(NGF)表达的上调,促使神经细胞修复。方法:大鼠脑出血(ICH)模型建立后随机分为Ea-BMSCs组(电针与细胞移植联合组)、BMSCs组(细胞移植组)、Control组(对照组),通过Western bloting方法检测各组在不同时间点BDNF、NGF总蛋白表达的变化。结果:与BMSCs组和Control组相比(除第1天外),Ea-BMSCs组大鼠ICH病灶周围NGF蛋白表达上调,差异有统计学意义(P <0. 05),且移植后的第3天、第5天、第14天BDNF蛋白表达显著增加,差异有统计学意义(P <0. 05)。结论:电针联合应用同种异体BMSCs移植后上调BDNF和NGF蛋白的表达可能是ICH神经元修复的分子基础之一。Objective In order to furlher investigate the mechanisnl of neuronal repair,we explore the effects of BDNF and NCF expression in intracerebral hemorrhage in rats after treatment of the electro - acupuncture combined with BMSCs transplan- tation. Method Injecting collagenase and heparin into caudate nucleus to prepare ICH model rats, which were randomly divided into Ea- BMSCs group, BMSCs group and Control group. The expression of BDNF, NGF protein were observed by RT -PCR and Western blot methods. Results Compared with BMSCs group and Control group (except ld outside) , NGF protein expres- sion of Ea - BMSCs gwup around ICH lesions in rats was up - regulated significantly ( P 〈 0. 05 ) ; BDNF protein expression was increased significantly at 3 days,5 day-s and 14 day-s (P 〈 0. 05 ). Conclusion Ea combined with BMSCs increased the ex- pression of BDNF and NGF protein,which may be one of the molecular basis of neuronal repair from ICH.
