摘  要:目的:研究IGF-1R基因对乳腺癌细胞株MDA-MB231细胞增殖及迁移能力的影响。方法:将本课题组前期构建的质粒p EGFP-N1-IGF-1R转染入MDA-MB231细胞系,采用Western blot法和Realtime PCR法检测IGF-1R蛋白与mRNA的表达情况;通过MTT法和Transwell法检测p EGFP-N1-IGF-1R对MDA-MB231细胞增殖及迁移侵袭能力的影响。结果:p EGFP-N1-IGF-1R转染入MDA-MB231细胞后,其IGF-1R的表达水平明显上调;与空白对照组和空载体组相比,过表达组细胞增殖能力明显增强(P <0. 05),细胞迁移能力增强(P <0. 05)。结论:IGF-1R基因能够增强MDA-MB231细胞的增殖能力及迁移能力。Objective: To explore the effects of p EGFP-N1-IGF-1 R on proliferation and migration of MDA-MB231 cells. Methods: The p EGFP-N1-IGF-1 R was transfected into MDA-MB231 cells. The expression of IGF-1 R gene was detected by Real-time PCR and Western blot. The effects of proliferation and migration of MDA-MB231 cells were examined by MTT and Transwell assay respectively. Results: The MDA-MB231 cells transfected with p EGFP-N1-IGF-1 R plasmid expressed a higher level of IGF-1 R mRNA and protein steadily. The capabilities of proliferation and migration of MDA-MB231 cells in p EGFP-N1-IGF-1 R/MDA-MB231 group were increased significantly compared with MDA-MB231 group and p EGFP-N1/MDA-MB231 group( P〈0. 05). Conclusion: IGF-1 R gene can enhance the abilities of proliferation and migration of MDA-MB231 cells.
