摘 要:目的探讨miR-34a在鼻咽癌中的表达及其临床意义。方法收集30例鼻咽癌组织标本,qRTPCR检测鼻咽癌组织中miR-34a表达量,对miR-34a表达与肿瘤临床病理特征及患者预后相关性进行分析,探讨miR-34a表达是否影响鼻咽癌细胞的迁移及侵袭能力。结果 30例鼻咽癌组织中miR-34a的相对表达量为(2.367±0.235),而对应癌旁组织为(5.399±0.463);其中22例鼻咽癌组织(73.33%)中miR-34a的相对表达量低于对应癌旁组织,差异有统计学意义(P<0.001);存在淋巴结转移及具有较高的TNM分期(Ⅲ+Ⅳ)的鼻咽癌组织中miR-34a的表达更低(P<0.01)。结论 miR-34a在鼻咽癌组织中表达下调,并与患者不良临床病理特征相关,上调miR-34a表达可能会抑制鼻咽癌细胞增殖、迁移和侵袭,促进凋亡,miR-34a基因可能作为一种抑癌基因在鼻咽癌的发生和发展中发挥重要作用。Objective To explore the expression of miR-34a in nasopharyngeal carcinoma and its clinical significance. Methods 30 nasopharyngeal carcinoma tissue samples were collected in the study.The expression quantity of miR-34a in these samples were detected by qRT-PCR.The correlations between expression of miR-34a and the clinicopathological features of tumors,and patients’ prognosis were analyzed to investigate whether miR-34a expression can affect the migration and invasion of nasopharyngeal carcinoma cells. Results The relative expression quantity of 30 nasopharyngeal carcinoma tissue was(2.367±0.235),while that of corresponding paracancerous tissue was(5.399±0.463);In 22 cases of nasopharyngeal carcinoma tissue samples(73.33%),the relative expression of miR-34a was lower than that of corresponding paracancerous tissues (P 〈0.001);Less expression of miR-34a was found in nasopharyngeal carcinoma tissue with lymph node metastasis and higher TNM stage(Ⅲ+Ⅳ) (P 〈0.01). Conclusion The expression of miR-34a in nasopharyngeal carcinoma is down-regulated,which is associated with poor clinical pathological features of patients;increasing the expression of miR-34a may restrain nasopharyngeal carcinoma cell from proliferation,moving and invasion,and accelerate its apoptosis at the meantime.MiR-34a gene may play an important role as a tumor suppressor gene in the occurrence and development of nasopharyngeal carcinoma.
