摘  要:目的在细胞水平上鉴定多肽-2(Peptide-2)与阿尔茨海默病β-分泌酶(BACE1)结合的特异性。方法合成经过二硫键修饰的Peptide-2,采用激光扫描共聚焦显微镜术及流式细胞术鉴定Peptide-2对表达BACE1的人源胚胎肾293(HEK293)细胞的结合特异性。结果激光扫描共聚焦显微镜显示Peptide-2在表达BACE1的HEK293细胞上有高富集荧光亮点(红色荧光),流式细胞术鉴定出Peptide-2与HEK293细胞中BACE1结合的荧光强度值为102. 352,PBS对照组的荧光强度值为16. 29,差异有统计学意义(P <0. 05)。结论 Peptide-2在细胞水平能特异结合至BACE1,为阿尔茨海默病β-分泌酶抑制剂的筛选提供了一个新的研究方向。Objective To identify the specific peptide-2 binding on the surface of β-secretase(BACE1) at cellular level,which was screened by phage display peptide library through preliminary experiment. Methods The peptide-2 was synthetized by chemical methods after modification by sulfur bonds. The specificity of peptide-2 binding to BACE1 on HEK293 cell was identified by confocal laser microscope and flow cytometry. Results The confocal laser scanning microscope identified that the peptide-2 could bind to BACE1 on HEK293 cell with high fluorescence(red). The fluorescence value of peptide-2 bind to BACE1 was 102. 352,and the fluorescence value of PBS control was 16. 29(P 0. 05) by flow cytometry. Conclusion The small molecular peptide named peptide-2 binding to BACE1 on HEK293 cell specifically could provide a new direction for screening β-secretase inhibitor candidate of Alzheimer's disease.
