摘  要:目的 探讨微小核糖核酸-379-5p(miR-379-5p)对膀胱癌细胞增殖、迁移和侵袭的影响.方法 通过实时荧光定量PCR (qRT-PCR)法检测膀胱癌组织细胞与正常组织细胞中miR-379-5p表达水平,分别将miR-379-5p mimics序列和miR-NC序列转染至人膀胱移行细胞癌细胞系um-uc-3,作为miR-379-5p mimics组和miR-NC组,通过Western blot实验检测转染效率,采用MMT实验和Transwell实验比较两组um-uc-3细胞增殖、迁移以及侵袭能力.结果 qRT-PCR检测结果显示,膀胱癌组织中miR-379-5p mRNA相对表达量为(0.52 ±0.38),显著低于正常组织中miR-379-5p mRNA相对表达量(1.00±0.51,P<0.05),um-uc-3中miR-379-5p mRNA相对表达量为(0.45±0.23),显著低于人膀胱上皮永生化细胞SV-HUC-1中miR-379-5p mRNA相对表达量(1.00±0.14,P<0.05);Western blot结果显示,miR-379-5p mimics组miR-379-5p表达量升高,转染有效;MTI实验显示,miR-379-5pmimics组细胞在不同时间点OD值均小于miRNC组;Transwell实验显示,miR-379-5p mimics组细胞迁移和侵袭数量均显著少于miR-NC组(P<0.05).结论 miR-379-5p在膀胱癌组织细胞中表达均下调,体外上调miR-379-5p表达可有效抑制膀胱癌细胞的增殖、迁移和侵袭能力.Objective To explore the effects of micro ribonucleic acid-379-5p (miR-379-5p) on the proliferation,migration and invasion of bladder cancer cells.Methods The expression level of miR-379-5p in bladder cancer tissues and normal tissues was detected by real-time quantitative PCR (qRT-PCR),and the miR-379-5p mimics sequence and miR-NC sequence were transfected into human bladder transitional cell carcinoma um-uc-3 cell and taken as the miR-379-5p mimics group and the miRNC group respectively.The transfection efficiency was detected by Westem blot,and the proliferation,migration and invasion of um-uc-3 cells were compared by MMT and Transwell experiments.Results The relative expression of miR-379-5p mRNA in the bladder cancer tissues was significantly lower than that in the normal tissues [(0.52 ± 0.38) vs.(1.00 ± 0.51),P 〈 0.05],and the relative expression of miR-379-5p mRNA in the um-uc-3 was significantly lower than that in human bladder epithelium immortalized cells SV-HUC-1 [(0.45 ± 0.23) vs.(1.00 ± 0.14),P 〈 0.05].The results of Western blot showed that the expression of miR-379-5p in miR-379-5p mimics group was increased and the transfection was effective.The MTT assay showed that the cells OD value at different time points in the miR-379-5p mimics group was lower than that in the miR-NC group.The Transwell experiments showed that the number of cell migration and invasion in the miR-379-5p mimics group was significantly less than that in the miR-NC group (P 〈 0.05).Conclusions The expression of miR-379-5p is down regulated in the bladder cancer tissues,and the external up-regulation of miR-379-5p expression can effectively inhibit the proliferation,migration and invasion of bladder cancer cells.
