摘 要:目的:探索mi R-939对非小细胞肺癌的侵袭能力及上皮细胞-间充质转化过程(epithelial-mesenchymal transition,EMT)的影响。方法:实时聚合酶链反应(q RT-PCR)检测mi R-939在非小细胞肺癌组织中的表达。Transwell侵袭实验分析mi R-939对细胞侵袭能力的影响。通过Western blot检测EMT相关标志物的表达水平。结果:与癌旁组织相比,mi R-939在NSCLC组织中的表达水平明显升高,且与TNM分期和淋巴结转移密切相关;同时,与正常肺上皮细胞相比,mi R-939在NSCLC细胞系中的表达也显著升高。下调mi R-939表达后,细胞侵袭能力明显降低(P<0.05)。另外,mi R-939下调后可使EMT相关标志物E-cadherin升高,Vimentin降低。结论:mi R-939可能作为不良因子,促进非小细胞肺癌侵袭转移,从而影响肿瘤发展。Objective:The purpose of this study was to examine the expression and biological role of mi R-939 in human non-small cell lung cancer(NSCLC)and epithelial-mesenchymal transition(EMT)progress. Methods:Quantitative real-time polymerase chain reaction(q RT-PCR)was used to detect the expression of mi R-939 in NSCLC. Transwell analysis was used to determine the effect of mi R-939 on cell invasion. EMT-related markers were detected by Western blot. Results:The expression of mi R-939 in NSCLC tissues was significantly higher than that in adjacent non-cancerous tissues,and closely related to TNM staging and lymph node metastasis.Meanwhile,compared with normal lung epithelial cells,the expression of mi R-939 in NSCLC cell lines was also significantly increased.The invasive ability of mi R-939 cells was obviously decreased after down-regulation of mi R-939 expression(P 0.05). In addition,mi R-939 down-regulation caused E-cadherin increasing and Vimentin decreasing. Conclusion:mi R-939 may serve as an adverse factor to promote cell invasion,thus affecting the development of NSCLC.
