摘  要:目的:评价X连锁凋亡抑制蛋白(XIAP)基因在大肠癌细胞系和组织中的表达,并将表达与患者的临床病理特征进行关联分析。方法:应用RT-PCR检测XIAP mRNA在大肠癌细胞系和大肠癌组织中的表达情况;应用免疫组化评价手术标本XIAP的表达情况。结果:XIAP mRNA在大肠癌细胞系(HT29,SW620和LoVo)中均较正常肠上皮细胞呈显著高表达;大肠癌组织中的XIAP表达量显著高于对应的无癌组织;高表达的XIAP与阳性淋巴结转移、较差的肿瘤分化情况和较差的病理分期均存在相关性(P<0.05)。结论:XIAP的表达情况可以作为大肠癌的诊断及预后标志物。Objective: The present study is to evaluated the expression of X-linked inhibitor of apoptosis protein(XIAP) in colorectal cancer(CRC), and associate the clinicopathologica factors to XIAP. Methods: RT-PCR was used to detect the expression between CRC and normal human intestinal epithelial cell lines(HIEC). Immunohisto chemistry was performed to evaluate the expression of XIAP in surgical specimens. Results: CRC cells(HT29,SW620 and LoVo showed significantly higher expression of XIAP to the HIEC) cell The expression. level of XIAP mRNA in CRC samples was significantly higher than that in corresponding non-tumorsamples Higher XIAP protein expression was significantly associated with TNM. staging, tumor differen tiation and lymph node metastasis(P〈0.05). Conclusion: The status of XIAP expression might become a CRC marker for diagnostic, and prognostic.
