摘  要:目的制备N-十八烷基壳聚糖膜,研究其体外凝血性能。方法以壳聚糖(chitosan,CS)和十八醛为原料,还原氨法制备不同取代度(40%、20%、10%)的N-十八烷基CS,流延法分别制备CS膜和N-烷基CS膜,红外、元素分析和溶胀率检测表征材料的结构、组成和吸水性,MTT法评价材料的细胞毒性,体外凝血时间研究材料的凝血性能。结果制备出不同取代度的N-十八烷基CS,并成功制备成膜。CS膜和N-十八烷基CS膜均无细胞毒性。不同取代度的N-十八烷基CS膜的体外凝血性能均优于纯CS膜,其凝血性能随十八烷基取代度的增加而增强。结论与纯CS膜相比,N-十八烷基CS膜具有促凝作用,且取代度越大,材料的凝血效果越好,表现出良好的生物相容性,有望作为医用敷料使用。Objective To prepare N-octadecyl chitosan films and study its coagulation performance in vitro. Methods Using chitosan(CS) and octadeealdehyde as raw materials, N-octadecyl CS with different degrees of substitution (DS 40% , 20% and 10% ) was prepared by reductive reaction. Its structure, composition and water absorbency properties were characterized by infrared, elemental analysis and swelling ratio detection. CS and N-octadeeyl CS solution was cast to form films. The cytotoxieity and coagulation properties of N-oetadecyl CS were evaluated by MTI" method and coagulation time in vitro respectively. Results N-octadecyl CS with different degrees of substitution and the corresponding film were successfully prepared. Both the CS membrane and the N-octadecyl CS film had no toxicity. The in vitro coagulation effect of any N-octadecyl CS film was better than that of CS film. Furthermore, the blood clotting effect was enhanced with the increase in DS. Conclusion Compared with CS films, N-octadecyl CS films have obvious procoagulant effect. The greater the DS is, the better the coagulation effect. Besides good biocompatibility, N-octadecyl chitosan can potentially be used as a safe wound dressing material.
