摘 要:目的检测婆罗双树样基因4(SALL4)基因在胶质瘤中的表达水平,探讨SALIA基因对胶质瘤细胞的增殖影响。方法生物信息手段分析癌症基冈组冈谱(TCGA)中SALL4基因的表达水平与预后的关系;定量聚合酶链反应(qPCR)验证SALL4基因在56例高级别胶质瘤(WHOⅢ、Ⅳ级)标本及6例非瘤脑组织标本中的表达差异川、干扰RNA(siRNA)沉默SALL4基网后,细胞计数试剂盒(CCK-8)试验、细胞周期试验检测肿瘤细胞的增殖变化;Westernblot法检测SALIA基因被沉默后SALL4和第10号染色体上缺失与张力蛋白同源的磷酸酯酶基因(PTEN)蛋白的表达变化。结果来源于TCGA的数据表明SALL4基因在肿瘤中高表达,约50%的SALL4低表达的胶质瘤母细胞瘤患者获得了更高的生存率;在56例高级别胶质瘤标本中SALL4的表达水平较6例非瘤脑组织更高(2.89±2.53比0.70±0.30,P=0.000);在SALL4基因被沉默后胶质瘤细胞的增殖能力被抑制,PTEN蛋白表达升高。结论SALL4基因参与了胶质瘤细胞生长的调控,抑制SALL4基因的表达能抑制胶质瘤细胞的增殖。Objective To investigate the expression of sal - like protein 4 (SALIA) gene in glioma and the effect of SALL4 gene on proliferation of glioma cells. Methods By bioinformatics, the data from The Cancer Genome Atlas (TCGA) were analyzed to explore the expression of SALIA gene and its relation to prognosis, qPCR was used to detect the expression of SALIA in 56 cases of grade ]lI and grade IV glioma samples according to the World Health Organization (WHO) classification system and 6 cases of nonminor brain tissue. Cell counting kit -8 (CCK -8) test and cell cycle test were performed to detect the proliferation of glioma cells after knocking down the SALIA gene. The protein expression of SALL4 and phosphatase and tensin homologue deleted on chromosome ten (PTEN) was detected by Western blotting after SALIA gene was silenced. Results Data from TCGA showed that SALL4 gene was highly expressed in tumors, and about 50% of patients with glioblastoma with low expression of SALIA had higher survival rate ; The expression of SALL4 in 56 high grade glioma samples was higher than that in 6 nontumor brain tissues (2.89 ±2. 53 vs. 0.70 ±0. 30), and its high expression had a relation to early survival rate. After the SALIA gene was silenced, the proliferation of glioma cells was inhibited, and the expression of FFEN was increased. Conclusion SALL4 gene is involved in the regulation of the growth of glioma cells. And inhibitin~ the exoression of SALIA gene can inhibit the proliferation of glioma cells.
