摘 要:目的:探讨miR-30c在胃癌中的表达及其对胃癌细胞MGC803增殖和侵袭等恶性生物学行为的影响。方法:采用逆转录定量PCR(RT-qPCR)法检测胃癌和癌旁组织中miR-30c的表达;将胃癌细胞MGC803进行转染,其中转染组细胞转染miR-30c-mimic,阴性对照组转染NC-mimic,正常对照组细胞不做任何处理,采用RT-qPCR法检测miR-30cmRNA的表达情况以判断转染效率,采用CCK8法测定细胞增殖情况,采用细胞划痕愈合试验和Transwell侵袭试验测定细胞迁移和侵袭能力。结果:与癌旁组织相比,在胃癌组织中miR-30c的表达量显著降低(P<0.05);转染组MGC803细胞miR-30cmRNA的表达量较正常对照组和阴性对照组显著增高(P<0.05),其细胞增殖率显著降低(P<0.05);细胞划痕距离显著升高(P<0.05),且细胞穿膜数量显著降低(P<0.05)。结论:miR-30c在胃癌组织中异常低表达,胃癌细胞MGC803中过表达miR-30c可抑制胃癌细胞的增殖、迁移和侵袭。Objective: To investigate the expression of miR-30c in gastric cancer tissues, and its effect on the malignant biological behaviors such as proliferation and invasion of gastric cancer cell line MGC803. Methods: The expression of miR-30c in gastric cancer and paracancerous tissues was detected by reverse transcription quantitative PCR (RT- qPCR). The gastric cancer cells MGC803 was transfected, and the transfected group was transfected with miR-30c-mimic, the negative control group was transfected with NC-mimic, and the normal control cells were not treated. The expression of miR-30c mRNA was detected by RT-qPCR to determine the transfection efficiency. Cell proliferation was measured by CCK8 method. Cell migration and invasion ability were determined by cell scratch healing assay and Transwell invasion assay. Results: Compared with paracancerous tissues, the expression of miR-30c was significantly decreased in gastric cancer tissues (P〈0.05). The expression level of miR-30c mRNA in the transfected group was significantly higher than that in the normal control group and the negative control group(P〈0.05) , and the cell proliferation rate was significantly decreased(P 〈0.05). The cell scratch distance was significantly increased(P〈0.05), and the number of cell transmembrane was significantly decreased (P〈0.05). Conclusion: miR-30c is abnormally low in gastric cancer tissues and overexpression of miR-30c in gastric cancer cells MGC803 can inhibit the proliferation, migration and invasion of gastric cancer cells.
