摘 要:目的探索胱硫醚β-合成酶(cystathioneβsynthase,CBS)844ins68基因多态性与新疆汉族乳腺癌的关系。方法收集新疆汉族150例乳腺癌患者(乳腺癌组)和150例健康体检者(对照组)外周静脉血,运用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)的方法检测CBS844ins68基因型和基因频率,同时用电化学发光免疫分析仪检测肿瘤标志物。结果 1.两组年龄分布差异无统计学意义,但是否绝经、癌胚抗原(carcinoembryonic antigen,CEA)、糖类抗原15-3(carcinoembryonic antigen15-3,CA15-3)的分布差异有统计学意义(P﹤0.05);2.乳腺癌组CBS844ins68位点基因型DD、DI、II分别为94.0%、6%、0;对照组CBS844ins68位点基因型DD、DI、II分别为96.7%、3.3%、0;乳腺癌等位基因频率D、I分别为97.0%、3.0%;对照组等位基因频率D、I分别为98.3%、1.7%,两组基因型和等位基因频率分布差异无统计学意义(P>0.05);3.Logistic回归分析显示年龄﹤50岁、已绝经、甲胎蛋白(alpha fetoprotein,AFP)AFP≥7.0、CEA≥3.4、CA15-3≥25可增加新疆汉族乳腺癌发病风险3-9倍。结论年龄小于50岁及已绝经、AFP、CEA、CA15-3水平增高是乳腺癌发病的危险因素,CBS844ins68基因多态性与新疆汉族乳腺癌发病无相关性。Objective To explore the correlation between Cystathione beta synthase (cystathione β synthase,CBS) 844ins68 gene polymorphism and breast cancer in Xinjiang Han populations. Methods A total of 150 breast cancer patients in Xinjiang Han populations (patient group) and 150 health examination people (control group) were selected for the study.The polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to analyze the genotype and gene frequency of CBS gene 844ins68; Tumor markers were examined by electrochemical luminescence assay. Results As the results showed that the age distribution between the two groups was not statistically different,but menopause,carcinoembryonic antigen (CEA) and carcinoembryonic antigen 15-3 (CA15-3) was statistically different. Genotype DD,DI,II of CBS844ins68 gen in the patient group was 94.0%,6%,0,while in the control group were 96.7%,3.3%,0,respectively.The allele frequency DI of CBS844ins68 gen in the patient group was 97.0%,3.0%,respectively; however,in the control group was 98.3%,1.7%.The overall frequency distribution between the two genotypes and the allele frequency were not statistically different (P〉0.05). Logistic regression analysis showed that less than 50 years old,menopause,alpha fetoprotein (AFP)≥7.0,CEA≥3.4 and CA15-3≥25 increased the risk of breast cancer in Xinjiang Han populations 3-9 times. Conclusion It can be concluded that less than 50 years old,menopause and increase of AFP,CEA,CA15-3 levels may correlates with breast cancer in Xinjiang Han populations.CBS844ins68 gene polymorphism has no correlation with breast cancer in Xinjiang Han populations.
