脂肪干细胞移植对脑缺血大鼠脑组织MAG和OMgp表达的影响

摘　　要：目的观察脂肪干细胞(ADSC)移植对脑缺血模型大鼠缺血侧脑组织中髓鞘相关糖蛋白(MAG)和少突胶质细胞髓鞘糖蛋白(OMgp)表达的影响。方法 SD大鼠分为假手术组、模型组和ADSC组。体外分离培养ADSC。模型组和ADSC组线栓法制作大鼠脑缺血模型,假手术组接受相似手术处理,但是不插入线栓。ADSC组于造模成功后经侧脑室注射ADSC悬液,模型组注射等量磷酸盐缓冲液。各组大鼠分别于4 d、7 d和14 d处死取材,采用免疫组织化学法和Western blot法检测大鼠缺血侧脑组织中MAG和OMgp表达情况。结果模型组和ADSC组各个时间点缺血侧脑组织中MAG和OMgp表达水平较假手术组明显升高(P <0. 05); ADSC组各个时间点缺血侧脑组织中MAG和OMgp表达水平较模型组明显降低(P <0. 05)。结论 ADSC移植促进大鼠脑缺血后神经功能的恢复,其机制可能与抑制MAG和OMgp表达有关。Objective To investigate the effect of adipose-derived stem cell（ADSC） transplanting on the expression of MAG and OMgp in rats with focal cerebral ischemia. Methods Rats were randomly divided into sham operation group, middle cerebral artery occlusion group and ADSCs-treated group, each group 18 rats. ADSCs were harvested and purified. Rat focal cerebral ischemia models were established by the suture method in model group and ADSC group. ADSCs were injected into the lateral ventricle of ADSC group rats and equal amount of PBS were injected into the lateral ventricle of model group rats. Before sacrifice and on 4 d, 7 d and 14 d after reperfusion, the expressions of MAG and OMgp were detected by immunohistochemistry and Western blot. Results The expressions of MAG and OMgp in model group and ADSC group were significantly higher than those in sham group at each time point （ P 〈0.05）. The expressions of MAG and OMgp in ADSC group were lower than model group at each time point （ P 〈0.05）. Conclusion ADSC transplanting can promote neuronal plasticity,which partly by inhibiting the expressions of MAG and OMgp.
