摘 要:目的研究Notch基因在口腔癌及癌前病变患者癌组织、癌旁组织中的表达情况。方法回顾性选取口腔鳞状细胞癌患者组织标本36例和癌前病变组织标本30例,均取得相对应的癌旁组织标本。检测Notch信号通路受体Notch1、Notch3基因mRNA表达水平,比较不同病变组织Notch1、Notch3基因mRNA表达水平及表达阳性率,分析Notch1、Notch3基因表达阳性率与口腔癌病理特征的关系。结果口腔癌癌组织Notch1、Notch3基因mRNA表达水平均显著高于癌前病变癌组织(P <0. 05);口腔癌癌旁组织Notch1、Notch3基因mRNA表达水平均略高于癌前病变癌旁组织,但差异无统计学意义(P> 0. 05)。口腔癌癌组织Notch1、Notch3基因表达阳性率75. 00%、72. 22%均显著高于癌前病变癌组织50. 00%、46. 67%(P <0. 05);口腔癌癌旁组织Notch1、Notch3基因表达阳性率均略高于癌前病变癌旁组织,但差异无统计学意义(P> 0. 05)。Notch1基因表达阳性率与口腔癌临床分期、淋巴结转移、病理分级有显著相关性(P <0. 05); Notch3基因表达阳性率与口腔癌临床分期、病理分级有显著相关性(P <0. 05),与淋巴结转移无显著相关(P> 0. 05)。结论口腔癌、癌前病变患者癌组织中Notch信号通路受体Notch1、Notch3基因mRNA表达水平较高,Notch基因尤其在口腔癌患者癌组织中表达活跃,且与口腔癌的发生发展具有相关性,但其在癌旁组织中的表达并不活跃。Objective To investigate the expression of Notch gene in cancer tissues and adjacent tissues of patients with oral cancer and precancerous lesions. Methods 36 cases of oral squamous cell carcinoma tissue specimens and 30 cases of precancerous lesion tissue samples were collected and the corresponding adjacent tissue specimens were also obtained. The expression levels of Notch signal pathway receptor Notch1 and Notch3 gene mRNA were calculated. The expression levels and positive expression rates of Notch1 and Notch3 gene mRNA in different tissues were compared. The relationship between positive expression rates of Notch1 and Notch3 genes and pathological features of oral cancer was analyzed. Results The expression levels of Notch1 and Notch3 gene mRNA were significantly higher in oral cancer tissues than precancerous lesion tissues ( P 〈0.05), and slightly higher in oral cancer adjacent tissues than precancerous lesion adjacent tissues ( P 〉0.05). The positive expression rates of Notch1 and Notch3 gene in oral cancer tissues were significantly higher than those in precancerous lesions (75.00% and 72.22% vs 50.00% and 46.67%) ( P 〈0.05). The positive expression rates of Notch1 and Notch3 gene in oral cancer adjacent tissues were slightly higher than those in the precancerous lesion adjacent tissues ( P 〉0.05). The positive expression rate of Notch1 was significantly correlated with clinical stage, lymph node metastasis and pathological grade of oral cancer ( P 〈0.05). The positive expression rate of Notch3 was significantly correlated with the clinical stage and pathological grade of oral cancer ( P 〈0.05), and the correlation with lymph node metastasis was not significant ( P 〉0.05). Conclusion The expression levels of Notch signal pathway receptor Notch1 and Notch3 genemRNA are higher in patients with oral cancer and precancerous lesions.
