摘 要:目的:分析miR-139在肺鳞癌组织中的表达及临床意义,预测与肺鳞癌预后相关的miR-139的靶基因。方法:从癌症基因组图谱(TCGA)数据库收集肺鳞癌数据集,下载患者临床特征、基因表达谱及预后随访信息。分析miR-139表达与肺鳞癌临床特征的相关性及对预后的影响。利用生物信息学方法预测miR-139的靶基因并进行生存分析。结果:在45对肺鳞癌和配对癌旁正常组织中,癌组织miR-139表达低于正常组织,且差异具有统计学意义(P <0. 001)。在Ⅰ/Ⅱ期肺鳞癌组织中,miR-139相对高表达组的总生存情况不如miR-139相对低表达组,其中位生存期分别为1713日和3253日,差异具有统计学意义(P <0. 001)。多因素Cox回归分析发现,miR-139是肺鳞癌的独立预后因子,高表达患者的死亡风险是低表达患者的2. 768倍(P <0. 001)。结论:在肺鳞癌组织中,miR-139相对高表达是一种预后不良因素,经独立验证之后有望作为预测肺鳞癌患者预后的生物标志物。Objective: To investigate the expression of miR-139 in squamous cell lung cancer(SCLC), and to predict possible target genes of miR-139 related to the prognosis of SCLC. Methogs: The gene expression profile in SCLC and their clinical data were downloaded from The Cancer Genome Atlas (TCGA). The prognostic value of miR-139 expression and the relationship between miR-138 expression and cnlinian fentures of SCLC ware analyzed. The target genes of miR-139 were predicted by online bioinformatic tools, and their relationship with prognosis were analyzed. Results: The expression of miR-139 in cancer tissue was significantly lower than that in the paired adjacent nontumorous tissue. In stage Ⅰ and Ⅱ cancer tissue, relatively higher expression of miR-139 was associated with inferior overall survival (OS). Furthermore, miR-139 was proved to be an independent negative prognosis factor for OS in multivariate survival analysis. Conclusion: miR-139 could be used as a potential prognostic marker in SCLC.
