摘  要:目的评价香菇多糖对SD大鼠母体是否有生殖毒性,对胚胎是否有致畸性,对其安全性进行毒理学评价。方法孕鼠随机分为阴性对照组、阳性对照组(0. 25 g/kg阿司匹林)、香菇多糖高剂量组、中剂量组、低剂量组(分别为8 g/kg BW、4 g/kg BW、2 g/kg BW,相当于推荐人用剂量的120倍、60倍、30倍)。于受孕第6 d~15 d经口给予受试物,并于第20 d处死。剖腹检查子宫连胎重、黄体数、早死胎数、晚死胎数、活胎数及着床数、胎仔性别、体质量、体长、外观有无异常。其后对胎仔进行骨骼检查及内脏检查。结果阴性对照组母体未见生殖毒性,胎仔有自发性的畸形。阳性对照组可见显著的母体生殖毒性及胎仔的致畸性。香菇多糖高剂量组、中剂量组、低剂量组对母体均无生殖毒性,胎仔有自发性的畸形,与阴性对照组比较,差异无统计学意义(P> 0. 05)。结论在本实验室条件下,香菇多糖对SD大鼠母体无生殖毒性,对胚胎无致畸性。Objective To evaluate whether the lentinan has reproductive toxicity to the SD rat,whether it is teratogenic to the embryo,and to perform toxicological evaluation of its safety. Methods Pregnant rats were randomly divided into 5 groups,including one negative control group,one positive control group(0. 25 g/kg Aspirin) and 3 lentinan groups(8 g/kg,4 g/kg,2 g/kg,equivalent to 120 times,60 times,30 times of the recommended human dose). Pregnant rats were administrated lentinan from the 6 thd to the 15 thd during pregnancy and were all killed at the 20 thd,and then the indicators for teratogenicity were examined,such as weight of uterine with fetal,number of implantation,corpus luteum,survived fetus,dead fetus,absorptive fetus of pregnant rats,gender,weight,body length,appearance malformation. Thereafter,bone and visceral malformation of fetal rats were examined. Results The negative control group showed no reproductive toxicity,and the fetus had spontaneous deformity. The positive control group showed significant maternal reproductive toxicity and teratogenicity of the fetus. The high,medium and low lentinan doses groups had no reproductive toxicity to the mother,and the fetus had spontaneous deformity.Compared with the negative control group,the differences were not statistically significant(P〈0. 05). Conclusion Maternal toxicity and teratogenic toxicity are not observed in rats exposed to lentinan in this study.
