摘  要:目的探讨妊娠糖尿病孕鼠胎盘中脂代谢相关因子雷帕霉素靶蛋白(mammaliantargetofrapamy.ein,mTOR)和脂质(Lipin)表达情况。方法妊娠大鼠分成实验组(即糖尿病组),干预组(糖尿病模型+雷帕霉素)及对照组(柠檬酸钠缓冲液)。于孕21d取出胎盘组织,通过免疫组化技术检测各组胎盘组织中p-mTOR、Lipin的定位表达;利用Western印迹技术检测胎盘组织中磷酸化雷帕霉素靶蛋白p-mTOR、mTOR、Lipin蛋白表达水平:利用RT-PCR技术检测胎盘组织中mT0R、Lipin的mRNAs表达水平。结果实验组中胎盘组织p-mTOR及Lipin水平均高于对照组,而干预组水平介于两者之间(P〈0.05);结论GDM孕鼠可能通过激活mTOR通路,使其过度磷酸化,提高Lipin水平,进而影响胎盘组织中脂质代谢。Objective To investigate the expression of lipid metabolism related factors (mTOR, Lipin) in placenta of gestational diabetes mellitus rats. Methods Pregnant rats were di- vided into experimental group (diabetic group), intervention group (diabetic model + rapamycin) and control group ( sodium citrate buffer) . On the 21 st day of gestation, the placenta tissues were taken out, and the expression of p-mTOR and Lipin were detected by immunohistochemistry, the expression of p-mTOR, roTOR and Lipin were detected by Western Blot, and the expression of mTOR and Lipin mRNAs were detected by RT-PCR. Results The levels of p-mTOR and Lipin in experimental group were higher than those in control group, but the levels in intervention group were between them (P 〈 0. 05 ) . Conclusion The abnormal lipid metabolism in the placenta of GDM pregnant rats may be caused by activating the mTOR pathway and increasing the level of Lip- lB.
