摘 要:目的研究SALL4在胃癌患者与健康人群血清中含量差异,并探讨SALL4分子在胃癌进展中的作用。方法选择2016年3月~2018年4月在成都市第三人民医院治疗的47例胃癌患者和参加体检的29例健康志愿者。采用酶联免疫吸附测定方法检测血清中SALL4含量;采用Real-time PCR和Western blot分析SALL4在多种胃癌细胞株和胃黏膜细胞株中的mRNA水平和蛋白水平表达情况;通过siRNA干扰研究SALL4对胃癌细胞迁移和侵袭能力的影响;Western blot分析上皮间质转化(EMT)相关分子表达情况。结果 SALL4在胃癌患者血清中的含量显著高于健康志愿者,差异有高度统计学意义(P <0.0001)。SALL4在多种胃癌细胞株中mRNA水平和蛋白水平的表达情况均高于胃黏膜细胞株。干扰SALL4表达后,胃癌细胞的迁移能力和侵袭能力均显著降低,差异有统计学意义(P <0.05)。SALL4表达降低可导致E-cadherin表达上调,snail、β-catenin和Wnt3a表达下调。结论SALL4与胃癌进展相关,可能通过诱导胃癌细胞EMT进程,促进细胞迁移和侵袭。Objective To investigate the difference of serum SALIA levels between gastric cancer patients and healthy people, and to explore the role of SALIA in the progression of gastric cancer. Methods A total of 47 patients with gastric cancer who were treated in the Third People's Hospital of Chengdu from March 2016 to April 2018 and 29 healthy volunteers who participated in the physical examination were selected. The serum levels of SALIA were detected by ELISA. The expression of SALIA in mRNA level and protein level in various gastric cancer cell lines and gastric mu- cosa cells were analyzed by Real-time PCR and Western blot respectively. The effects of SALIA on the cell migration and invasion ability were analyzed by siRNA interference. The expression levels of related molecules of epithelial-mesenchymal transition (EMT) were detected by Western blot. Results The serum SALIA levels in patients with gastric cancer were significantly higher than those in healthy volunteers, and the difference was highly statistically significant (P 〈 0.0001). The expression of SALIA mRNA and protein in various gastric cancer cell lines was higher than that in gastric mu- cosal cell lines. The migration and invasion ability of gastric cancer cells were significantly decreased after interfering SALIA expression, and the differences were statistically significant (P 〈 0.05). Decreased expression of SALIA leaded to the up-regulation of E-cadherin expression and the down-regulation of snail, β-catenin and Wnt3a. Con- clusion SALIA is related to the progression of gastric cancer. It may promote cell migration and invasion by inducing the EMT process in gastric cancer cells.
