高表达p27基因对子宫颈癌HeLa细胞增殖的影响研究

摘　　要：目的:探讨高表达p27基因对子宫颈癌HeLa细胞增殖的影响。方法:利用PT-PCR技术从宫颈癌HeLaRNA中扩增p27cDNA,利用脂质体法转染子宫颈癌HeLa细胞,并利用Western blot法、RT-PCR法对p27基因高表达细胞株进行筛选,并利用流式细胞术、MTT法对p27基因高表达对细胞增殖的影响情况进行检测。结果:研究成功获取p27高表达HeLa细胞株,且研究结果显示p27基因高表达可在一定程度上对细胞增殖产生抑制功效,并可促使G1期细胞数显著增加。结论:HeLa细胞中p27基因高表达可在一定程度上对细胞增殖现象进行抑制,且p27基因可作为子宫颈癌基因治疗的靶基因。Objective：To explore the effect of overexpression p27 gene on the proliferation of HeLa cells incervical cancer.Methods：The p27cDNA was amplified from HeLaRNA of cervical cancer by PT - PCR. The HeLacells were transfected with liposome method , and the overexpression cell line of p27 gene was screened by Westernblot and RT - PCR,and the effect of overexpression of p27 gene on cell proliferation was detected by flow cytometryand MTT method.Result：In this study, hela cell lines with overexpression of p27 were successfully obtained. Theresults showed that the overexpression of p27 gene could inhibit cell proliferation to a certain extent and significantlyincrease the number of cells in G1 phase.Conclusion：The overexpression of p27 gene in HeLa cells can inhibit cellproliferation to a certain extent, and p27 gene can be used as the target gene for cervical cancer gene therapy.
