摘 要:【目的】目前羊肚菌栽培菌种品性退化,羊肚菌种质资源的筛选评价对于选育优质新品质具有重要意义。【方法】本研究采用核糖体DNA转录间隔区(Internal Transcribed Spacer,ITS)序列分析技术,对收集到的18个羊肚菌菌株进行菌株鉴定和系统发育分析,同时应用ISSR(inter-simple sequence repeat)分子标记手段,进行遗传多样性以及亲缘关系分析。【结果】10个菌株为梯棱羊肚菌(Morchella importuna),2个为六妹羊肚菌(Morchella sextelata),4个为高羊肚菌(Morchella elata),2个野生羊肚菌为粗柄羊肚菌(Morchella crassipes)。ISSR聚类分析显示出供试菌株的总体相似性为0.52,相似系数为0.54时聚为两大类群。【结论】18株菌株间具有明显的遗传多样性。【Objective】Presently,due to the degradation of the quality and agronomic traits of cultivation Morchella strains,screening and evaluating of Morchella strain have a great significance for breeding high quality varieties.【Method】ITS sequence analysis was applied to identify the 18 Morchella strains,and ISSR analysis was used to explore the genetic diversity of the 18 Morchella samples.【Result】10 strains were identified as Morchella importuna,2 strains were identified as Morchella sextelata,4 strains were identified as Morchella elata,and 2 strains were identified as Morchella crassipes.The cluster map showed that the overall similarity of tested strains was 0.52,and the tested strains were clustered into two groups at similarity coefficient of 0.54.【Conclusion】Morchella strains have a distinct genetic diversity.
