摘  要:目的:从突触可塑性和轴突运输探究坐骨神经损伤大鼠模型痛觉过敏的机制。方法:采用坐骨神经慢性压迫性损伤(chronic compression injury,CCI)诱导大鼠外周神经性疼痛,检测CCI后大鼠机械性痛觉过敏、冷触诱发痛以及热痛觉过敏的指标。实时定量PCR检测CCI后14 d及21 d的大鼠同侧L4-6背根神经节中GAP-43、NCAM、PSD-95、Syp、kinesin和dynein的表达。结果:CCI术后14 d大鼠的机械性痛觉过敏、冷触诱发痛以及热痛觉过敏指标均降低(P均<0. 01)。CCI组术后14 d GAP-43和NCAM的表达增加(P均<0. 01),PSD-95、Syp、kinesin和dynein的表达均降低(P <0. 05)。术后21 d GAP-43和NCAM的表达均下降(P <0. 01,P <0. 05),PSD-95、Syp、kinesin和dynein的表达均增加(P <0. 05)。结论:坐骨神经损伤14 d内,主要以修复神经元结构为主。而损伤14 d后,兴奋的传递以及轴突运输启动。Objective: To explore the mechanism of hyperalgesia with sciatic nerve injury rats model in terms of synaptic plasticity and axonal transport. Methods: Peripheral neuropathic pain in rats were induced by chronic compression injury (CCI) of sciatic nerve.The indexes of mechanical hyperalgesia,cold allodynia and thermal hyperalgesia in rats after CCI were detected.The expressions of GAP-43,NCAM,PSD-95,Syp,kinesin and dynein in the L4-6 dorsal root ganglion at 14 d and 21 d after CCI were detected by real-time quantitative PCR detection. Results: The indexes of mechanical hyperalgesia,cold allodynia and thermal hyperalgesia in rats in 14 d after CCI were reduced ( P 〈0.01).The expressions of GAP-43 and NCAM in 14 d after CCI were increased ( P 〈0.01),and the expressions of PSD-95,Syp,kinesin and dynein were reduced ( P 〈0.05).The expressions of GAP-43 and NCAM in 21 d after CCI were reduced ( P 〈0.01 or P 〈0.05),and the expressions of PSD-95,Syp,kinesin and dynein were increased ( P 〈0.05). Conclusion: The repair of the neuron structure is predominant in the 14 d after sciatic nerve injury.After 14 d of injury,transmission of excitement and axon transport are initiated.
