人甲状腺乳头状癌中微小RNA-381-3p低表达及其临床意义探究

摘　　要：目的探究人microRNA-381-3p(miR-381-3p)在甲状腺乳头状癌(PTC)中的表达并探讨其临床意义。方法收集从2017年02月到2018年02月在泰山医学院附属医院甲状腺外科手术的129例甲状腺乳头状癌(PTC)患者的癌组织和癌旁组织,其中男56例,女73例,年龄24~85岁,平均44. 7岁。提取各组织中的微小RNA,应用实时荧光定量聚合酶链反应(RT-qPCR)方法检miR-381-3p的表达情况。结果甲状腺乳头状癌组织中miR-381-3p的相对表达量较癌旁组织明显下调[4. 595±1. 630 Vs 2. 929±1. 198,P=0. 000]。通过对年龄、性别、肿瘤大小、是否腺外侵犯等进行因素分层后的组间比较显示miR-381-3p的相对表达量与PTC患者性别、年龄无关,而与肿瘤直径、腺外侵犯呈有关。结论 miR-381-3p在PTC中的表达异常下调,可能与PTC的发生、侵袭和转移相关。Objective：To investigate the abnormal expression of miR-381-3p in the patients with papillary thyroid carcinoma, and to explore the clinical significance of it. Methods： 129 cancer tissues and adjacent normal tissues from patients with papillary thyroid carcinoma were collected in our hospital from February 2017 to February 2018, including 56 males and 73 females, and the mean age was 44. 7 years. MicroRNAs were extracted from the tissues, the expression of miR-381- 3p was determined by reverse transcription quantitative real-time PCR （RT-qPCR）. Results ： The expression level of miR- 381-3p was significantly clown regulated in cancer tissues than that in normal tissues 14. 595 ± 1. 630Vs2. 929 ± 1. 198, P = 0. 000 ]. The relative expression of miR-381-3p was negatively with tumor diameter, lymphatic metastasis and extrathyroid extension, but no correlation was found between the level of mir-381-3p with gender and age. Conclusion： The abnormal clown-regulated expression of miR-381-3p in PTC might be associated with the occurrence, invasion and metastasis of PTC.
