摘　　要：【目的】,为了获得凝结芽孢杆菌高产活菌的最优发酵条件和碳氮源。【方法】利用单因素试验的方法,对凝结芽孢杆菌培养基中碳源、氮源以及培养条件中温度、初始pH值等因素进行优化。【结果】建立最佳培养基为:无水葡萄糖20g/L,K_2HPO_4·3H_2O 1g/L,MgSO_4·7H_2O 0.2g/L,MnSO4·H2O 0.02g/L,玉米浆粉17g/L,最佳培养条件为:温度40℃,pH 5.5,摇瓶装液量150mL/500mL,摇床转速200r/min,培养时间20h。【结论】获得凝结芽孢杆菌高产活菌的最优发酵条件和碳氮源,筛选出培养凝结芽孢杆菌的优质低价原料,为下一步规模化发酵提供了依据。【Objective】The objective of this study was to optimize the fermentation conditions and sources of carbon and nitrogen of Bacillus coagulans producing viable bacteria, as well as the temperature and initial PH in culture medium.【Methods】 The fermentation conditions of Bacillus coagulans were optimized by single factor experiment.【Results】The optimal medium formulation was 20 g / L of anhydrous glucose, 1 g / L of K 2 HPO 4 · 3 H 2 O, 0.2 g / L of MgSO 4 · 7 H 2 O, 0.02 g / L of MnSO 4 · H 2 O and 17 g / L of corn syrup. Fermentation conditions： temperature 40 ℃, pH 5.5, shake flask liquid volume 150 mL / 500 mL, shaker speed 200 r / min, culture time 20 h.【Conclusion】The optimal fermentation conditions and carbon and nitrogen sources of higher viable bacteria of Bacillus coagulans were obtained, and the high quality and low price raw materials for Bacillus coagulans were screened to provide basis for the next large-scale fermentation.
