摘 要:以老山芹为原料,确定老山芹总黄酮微波辅助提取最佳工艺条件,并对其抗氧化活性及组成成分进行测定。在单因素试验的基础上,以老山芹总黄酮提取率为指标,通过响应面优化提取工艺,得到老山芹黄酮最佳的提取工艺为:微波功率290 W、料液比1:30 (g/mL)、微波时间740 s、乙醇浓度80%。在此条件下,老山芹总黄酮提取率达2.49%。高效液相色谱法(High performance liquid chromatography,HPLC)分析老山芹黄酮成分结果表明:老山芹黄酮中芦丁含量较高,质量分数为0.1521%;其次为芹菜素,质量分数为0.1072%;山奈酚和槲皮素质量分数分别为0.0598%、0.0301%。此外,以Vc为对照对老山芹黄酮进行抗氧化活性测定,结果表明:老山芹黄酮对O2-?、ABTS+?和DPPH自由基清除能力较强,其IC50值分别为1.066、0.219、0.339mg/mL,且有较强的还原力,表明老山芹黄酮有较强的抗氧化活性,可作为天然抗氧化剂的开发来源。In this study, the optimum conditions of microwave assisted extraction of total flavonoids from Heraclum dissectum were determined, also include their antioxidant activity and composition. On the basis of single-factor tests, with the extraction rate of total flavonoids from Heraclum dissectum as an indicator,the optimum extraction technology of flavonoids was obtained through response surface optimization:microwave power 290 W, the ratio of material to liquid was 1:30(g/mL), microwave time 740 s, ethanol concentration 80%. Under this condition, the extraction rate of total flavonoids from Heraclum dissectum reached 2.49%. The results of high performance liquid chromatography(HPLC) analysis showed that the content of rutin in flavonoids from Heraclum dissectum was high, the mass fraction was 0.1521%, followedby apigenin, and the mass fraction was 0.1072%. The mass fraction of kaempferol and quercetin were0.0598% and 0.0301%, respectively. In addition, the antioxidant activity of flavonoids was determined with Vc as a positive control. The results showed that the flavonoids from Heraclum dissectum had strong scavenging activities for superoxide anion radicals, ABTS radicals and DPPH radicals, the IC50 values were 1.066, 0.219, 0.339 mg/mL, respectively, and the reducing power was strong too, indicating that flavonoids from Heraclum dissectum have strong antioxidant activity and can be used as a source of natural antioxidant development.
