摘 要:目的:检测釉基质蛋白(EMD)诱导乳牙生理性根吸收不同时期的牙周膜干细胞(PDLSCs)向成牙骨质细胞方向分化的能力及矿化相关蛋白表达水平的变化。方法:选取根吸收早期(E组)、中晚期(M组)的乳牙和恒前磨牙(P组)作为组织来源,并分离培养其PDLSCs;然后再将各组PDLSCs分为2个亚组:实验组用含100 mg/mL EMD的DMEM诱导培养,对照组用DMEM培养;并于诱导7 d后检测各组细胞中CAP、CEMP-1及BSP、OCN、ALP、COL-1表达水平的差异。结果:各组细胞经EMD诱导培养7 d后,实时定量PCR检测结果显示,与对照组相比,各实验组PDLSCs中BSP、ALP、COL-1的表达水平均显著上调(P <0. 05),升高幅度为P组> E组> M组; P、E、M各实验组PDLSCs中CAP、CEMP-1 mRNA的表达水平为P组> E、M组(P <0. 05),E组与M组间无统计学差异(P> 0. 05); BSP、OCN的表达水平均为P组> E组> M组(P <0. 05); ALP的表达水平为P组> E、M组(P <0. 05),E组和M组间无统计学差异(P> 0. 05); COL-1的表达水平在各实验组间相比无统计学差异(P> 0. 05)。免疫组化染色结果显示,P、E、M各实验组PDLSCs中BSP、OCN、ALP、COL-1均呈阳性表达,对照组则显示染色不显著。结论:EMD可诱导乳牙PDLSCs向成牙骨质细胞方向分化。随着乳牙生理性根吸收的进行,乳牙PDLSCs向成牙骨质细胞方向分化的能力及矿化相关蛋白的表达水平均逐渐降低。AIM: To study the expression level of mineralization related protein in differentiation of periodontal ligament stem cells(PDLSCs) to cementoblasts during different physiological root resorption of the deciduous teeth induced by enamel matrix protein(EMD). METHODS: PDLSCs were isolated from deciduous teeth at early root absorption stage(group E), middle and late root absorption stage (group M) stages respectively as the tissue source, and those from permanent premolars(group P) were used as the controls. The expression of CAP, CEMP-1, BSP, OCN, ALP and COL-1 were detected after induced by EMD for 7 days. RESULTS: Group P showed the highest level of CAP and CEMP-1 mRNA expression( P 〈0.05). For BSP and OCN mRNA expression level, group P〉E〉M( P 〈0.05);for ALP expression group P〉E and M( P 〈0.05, E vs M, P 〉0.05). BSP, ALP and COL-1 expression level increased by EMD( P 〈0.05),the increase of group P〉group E〉group M. CONCLUSION: EMD may promote the differentiation of PDLSCs to cementoblasts. With the absorption of the roots of deciduous teeth, the ability of the differentiation of PDLSCs to cementobalsts and the level of the mineralization related protein secretion gradually decreased.
