摘　　要：目的:探讨膜联蛋白A5(ANXA5)对人胆管癌细胞QBC939增殖、凋亡、迁移和侵袭的影响。方法:构建3个沉默QBC939细胞ANXA5的shRNA质粒(ANXA5-sh1/2/3)和1个阴性对照质粒,慢病毒包装质粒后感染QBC939细胞,流式细胞仪检测感染效率。病毒感染QBC939细胞后,Western blotting检测细胞的ANXA5蛋白表达情况;流式细胞仪检测细胞周期和凋亡率变化;划痕实验和Transwell实验评估QBC939细胞迁移和侵袭能力的变化。结果:干扰组(KD)细胞中的ANXA5蛋白水平显著低于空白组(BC)和阴性对照组(NC)细胞中的ANXA5蛋白水平。与NC组和BC组细胞相比,KD组细胞中G0/1细胞的百分比和凋亡率增高,而细胞增殖活性降低。KD组细胞中的细胞迁移和侵袭能力也低于NC和BC组细胞。结论:RNA干扰QBC939细胞的ANXA5表达后,明显抑制其增殖、侵袭和诱导凋亡的作用。Objective： To investigate the effect of Annexin A5 （ANXA5） on proliferation, migration,invasion and apoptosis of human cholangiocarcinoma cell line QBC939. Methods： 3 shRNAANXA5 silencing plasmids （ANXA5-sh1/2/3） and 1 negative control plasmid were constructed to infecthuman bile duct carcinoma cell line QBC939. The infection efficiency, expression of ANXA5,apoptosis rate and cell cycle of QBC939 cell were measured separately. Results： Western blottingshowed that expressed ANXA5 protein in KD group cell（QBC939 cell with RNAi on ANXA5） was significantlylower than BC group（QBC939 cell） and NC group cells（QBC939 cell with scramble plasmid）.The percentage of G0/1 cells and apoptosis rate increased, migration, proliferation activity the invasionability decreased in KD group cells compared with NC and BC group cells. Conclusions：ANXA5 play important role in the migration and metastasis of cholangiocarcinoma cells. Inhibiting theexpression of ANXA5 significantly increase the apoptosis of QBC939 cells, reduce the proliferation,migration and invasion of QBC939 cells.
