摘 要:[目的]研究黄连解毒汤对健康大鼠肠道菌群结构的影响。[方法]雄性Wistar大鼠20只依据体重分为空白对照组和黄连解毒汤组,每组各10只。黄连解毒汤组按照700mg/kg的剂量给予黄连解毒汤提取物溶液灌胃,空白对照组大鼠给予相同体积的0.9%氯化钠溶液灌胃,两组均连续给药7d。7d后采用Illumina高通量测序技术检测两组大鼠的肠道菌群,寻找其中的差异菌群并对其进行生物学分析。[结果]在门水平菌群变化中,与空白对照组比较,服用黄连解毒汤临床剂量7d可以显著降低健康大鼠肠道中放线菌门和厚壁菌门细菌所占总菌的比例,两组差异具有统计学意义(P<0.05)。在属水平上,可以显著性降低健康大鼠肠道中Adlercreutzia菌、消化链球菌和克里斯滕森菌所占的比例,两组差异具有统计学意义(P<0.05,P<0.01)。[结论]通过肠道菌群实验发现,黄连解毒汤临床剂量服用7d后可对健康大鼠的肠道菌群产生一定的调节作用。[Objective]To study the effect of Huanglianjiedu Decoction on intestinal flora structure in healthy rats. [Methods] Twenty Wistar rats were divided into blank control group and Huanglianjiedu Decoction group, 10 of each group. Illumina high-throughput sequencing method was used to detect the intestinal flora of healthy rats and the rats after Huanglianjiedu Decoction administration(clinical dose) for seven days. The differential flora was identified and biological analysis was carried out. [Results] At the level of the phylum, compared with blank control group, taking the clinical dose of Huanglianjiedu Decoction for 7 days can significantly reduce the proportion of Actinobacteria and Firmicutes bacteria in the intestinal tract of Huanglianjiedu Decoction group rats. The difference between the two groups was statistically significant(P〈0.05). At the genus level, the proportion of Adlercreutzia, Peptococcaceae and Christensenellaceae in the intestinal tract of healthy rats was significantly reduced(P〈0.05, P〈0.01). [Conclusion]Illumina's high-throughput sequencing technology was sensitive and accurate. It is found through the intestinal flora experiment that the clinical dose of Huanglianjiedu Decoction can regulate the intestinal flora of healthy rats after seven days. It indicated that Huanglianjiedu Decoction has a good regulation effect on intestinal flora at a certain dose.
