摘 要:目的对丝素蛋白/氧化石墨稀复合膜复合材料的制备及在长骨骨不连中的治疗效果进行研究。方法分别制备丝素溶液和石墨烯溶液,以两种溶液为原始材料合成丝素蛋白/氧化石墨稀复合膜,通过扫描电镜观察其形态结构,将0.1wt%、0.2wt%、0.5wt%和1.0wt%浓度氧化石墨烯含量的丝素蛋白/氧化石墨稀复合膜与小鼠胚胎成骨细胞MC3T3-E1进行复合培养,采用MTT比色法在1、3、5和7d四个监测点对小鼠胚胎成骨细胞MC3T3-E1的增殖情况进行检测;用30只新西兰大白兔建立桡骨缺损性骨不连模型,随机分为15例观察组和15例对照组,观察组采用丝素蛋白/氧化石墨稀复合膜复合材料治疗,对照组只缝合伤口不行内固定,在治疗后4、8和12周行大体、组织学、生物力学测试和X射线评价骨缺损修复情况。结果 MC3T3-E1细胞的增值率随着氧化石墨烯浓度的增加逐渐降低(P<0.05),1、3、5和7d四个时点的细胞增值率逐渐增加,但差异无统计学意义(P>0.05);术后12周两组大白兔愈合界面骨组织标本的生物力学检测显示,观察组失效负荷、刚度均显著高于对照组(P<0.05),术后第4、8和12周观察组骨折愈合评分均显著高于对照组(P<0.05)。结论丝素蛋白/氧化石墨稀复合膜的相容性和生物力学性良好,在长骨骨不连的治疗中使用该方法利于早期进行骨愈合。Objective To preparation of silk fibroin/oxidized graphite thin composite membrane composites and to observation the effect of treatment in long bone nonunion. Methods The silk fibroin/oxidized graphite composite film was prepared by 2 kinds of solution as raw materials. The morphological structure was observed by scanning electron microscopy.0.1 wt%,0.2 wt%,0.5 wt% 1.0% by weight of graphene content of silk fibroin/oxidized graphite composite membrane and mouse embryonic osteoblasts MC3 T3-E1 were cultured in vitro,using MTT colorimetric method at 1 st,3 rd,5 th and 7 th day four monitoring points. The expression of MC3 T3-E1 in mouse embryonic osteoblasts was detected. 30 New Zealand rabbits were randomly divided into 15 observation groups and 15 control groups. The observation group was treated with silk fibroin/oxidized graphite thin composite membrane composite treatment,the control group accepted suture wounds but no fix inside. 4,8 and 12 weeks after treatment in general,histological,biomechanical tests,X-ray evaluation of bone defect repair. Results The increment rate of MC3 T3-E1 cells decreased gradually with the increase of graphene concentration.The difference was significant(P〈0.05),and the cell rate increased at 1 st,3 rd,5 th and 7 th day,but the difference was not significant(P〈0.05). The biomechanical test showed that the failure load and stiffness of the observation group were significantly higher than those of the control group at 12 weeks postoperatively. The difference was significant(P〉0.05). 4,8 and 12 weeks week after treatment,the observation group were significantly higher than the control group,the difference was statistically significant(P〈0.05). Conclusion The silk fibroin/oxidized graphite thin composite membrane compatibility and biomechanical good. In the treatment of long bone nonunion using this method is conducive to early bone healing.
