摘 要:目的探讨桑黄素对子宫内膜癌AN3-CA细胞凋亡和侵袭的影响。方法 MTT法分析不同浓度(10、20、40μmol/L)和不同作用时间(24、48 h)的桑黄素对AN3-CA细胞活性的影响;流式细胞学术分析不同浓度桑黄素对AN3-CA细胞凋亡的影响; Transwell实验分析不同浓度桑黄素对AN3-CA细胞侵袭的影响; Western blotting分析不同浓度桑黄素对AN3-CA细胞基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶-9(MMP-9)表达的影响;结果不同浓度的桑黄素作用于AN3-C细胞,24 h细胞活性高于同浓度48 h的细胞活性(P <0. 05)。AN3-CA细胞的凋亡率随着桑黄素浓度的升高而增加,侵袭能力随着桑黄素浓度的升高而减低(P <0. 05)。AN3-CA细胞中MMP-2和MMP-9蛋白表达随着桑黄素浓度的升高而下降(P <0. 05)。结论桑黄素可通过抑制MMP-2和MMP-9蛋白的表达影响子宫内膜癌AN3-CA细胞凋亡和侵袭。Objective To investigate effects of Morin on apoptosis and invasion of AN3-CA cells in patients with endometrial cancer. Methods Effects of different concentrations (10, 20 and 40 μmol/L) and different action times (24 h and 48 h) of Morin on activity of AN3-CA cells were analyzed by MTT assay. Flow cytometry was used to analyze effects of different concentrations of Morin on apoptosis of AN3-CA cells. Transwell assay was used to analyze effects of different concentrations of Morin on invasion of AN3-CA cells. Western blotting was used to analyze effects of different concentrations of Morin on expressions of matrix metalloproteinase-2 (MMP-2) and MMP-9 proteins in AN3-CA cells. Results In AN3-C cells affecting by different concentrations of Morin, activity at 24 h action was significantly higher than that at 48 h action at the same concentration ( P 〈0.05). Apoptosis rates of AN3-CA cells were increased with increasing Morin concentrations, while invasion abilities of AN3-CA cells were decreased with increasing Morin concentrations ( P 〈0.05). Expressions of MMP-2 and MMP-9 proteins in AN3-CA cells were decreased with increasing Morin concentrations ( P 〈0.05). Conclusion Morin may affect apoptosis and invasion of AN3-CA cells in patients with endometrial cancer by inhibiting expressions of MMP-2 and MMP-9.
