摘 要:目的:调查河南新乡地区汉族人群27个Y-STR基因座(包含3个双拷贝基因座)的遗传多态性,为法医学个体识别及法医DNA数据库的建立提供数据。方法:采集263名河南新乡地区汉族无关男性个体血样本,提取基因组DNA,采用AGCU Data Y30六色荧光试剂盒复合扩增,GeneMapper~ID X软件分析结果并分型。结果:DYS392、DYS389Ⅱ、DYS447、DYS522、DYS19、DYS448、DYS481、DYS460、DYS458、DYS635、DYS444、DYS643、DYS557等13个单拷贝Y-STR基因座的基因多样性(GD)大于0.7,DYS527 a/b和DYS385 a/b两个双拷贝Y-STR基因座的GD值大于0.8。在263名无关男性样本中,共检测到261种单体型,其中259种单体型仅出现1次,2种出现2次。27个Y-STR基因座的个体识别力和单体型多样性分别为0.996 1和0.999 9,单体型辨别能力为0.992 4。结论:27个Y-STR基因座组成的单体型在河南汉族群体中具有较高的遗传多态性。Aim: To investigate genetic polymorphism of 27 Y-STR loci( including three double-copy loci) among Han population in Xinxiang area,Henan province so as to provide basic data for personal identification,paternity identification and forensic DNA database. Methods: The blood samples of 263 unrelated males were collected for extracting genomic DNA. Amplification of 27 Y-STR loci was performed with AGCU Data Y30 6-dye kit,and Gene MapperID X Software was used for genotyping. Results: The gene diversity of 13 Y-STR loci ( DYS392,DYS389 Ⅱ,DYS447, DYS522,DYS19, DYS448, DYS481, DYS460, DYS458, DYS635, DYS444, DYS643, DYS557) was higher than 0. 7,and that of two double-copy Y-STR loci,DYS527 a/b and DYS385 a/b was above 0. 8. A total of 261 haplotypes were found,among which,259 haplotypes were observed only once,2 were observed twice,and the discrimination power,the haplotype diversity and the discrimination capacity were 0. 996 1,0. 999 9 and 0. 992 4,respectively. Conclusion: The haplotypes of 27 YSTR of Han population in Henan have abundant genetic polymorphism.
