吡格列酮调控离体新生大鼠心肌细胞增殖与肥大的研究

摘  要:目的研究吡格列酮与原代培养的新生大鼠心肌细胞(neonatal rat cardiac myocytes,NRCM)增殖和肥大之间的关系,探索出生后影响心肌细胞增殖与肥大的机制和临床治疗的潜在新靶点。方法 (1)NRCM的提取与培养;(2)NRCM加入吡格列酮处理;(3)利用免疫荧光染色检测NRCM增殖能力和细胞大小的变化;(4)数据分析(主要的统计学方法是非配对t检验和χ~2检验,采用SPSS 20,GraphPad和Image J统计软件进行统计分析,以P <0.05表示差异具有统计学意义)。结果 (1)吡格列酮组EDU阳性NRCM比例高于对照组,差异具有统计学意义(P=0.000 6);(2)吡格列酮组Ki-67阳性NRCM比例高于对照组,差异具有统计学意义(P=0.034 7);(3)吡格列酮组NRCM体积大于对照组,差异具有统计学意义(P=0.008 5)。结论吡格列酮促进离体新生大鼠心肌细胞增殖与肥大,可能与心脏的生理性肥大相关,本研究可能为病理性心脏肥大的治疗提供新的研究方向。Objective The main purpose of this study is through study the relationship between pioglitazone and the neonatal rat cardiac myocytes proliferation and hypertrophy to explore the mechanism of affect cardiomyocyte proliferation and hypertrophy after birth and new potential targets of clinical treatment. Methods (1) Isolation and culture of NRCM; (2) NRCM was treated with pioglitazone; (3) Changes in NRCM proliferation and cell volume were detected by immunofluorescence staining; (4)Statistical analysis (The main statistical methods are t test and χ^2 test, using SPSS 20, GraphPad and Image J statistical software for statistical analysis and P 〈 0.05 shows that the difference was statistically signifcant). Results (1) The proportion of EDU positive NRCM was significantly higher in pioglitazone group than in the control group, the difference was statistically signifcant (P=0.000 6); (2) The ratio of ki-67 positive NRCM in pioglitazone group was signifcantly higher than that in the control group, the difference was statistically significant (P=0.034 7); (3) The NRCM volume of pioglitazone group was signifcantly higher than that of control group, the difference was statistically signifcant (P=0.008 5). Conclusion Pioglitazone promoted the neonatal rat cardiac myocytes proliferation and hypertrophy. So pioglitazone may be associated with the physiologic hypertrophy of the heart. This study may provide a new research direction for the treatment of pathological myocardial hypertrophy.
