摘  要:目的利用紫杉醇诱导荷瘤兔VX2肿瘤细胞发生凋亡,行^(18)F-2-(5-氟-戊基)-2-甲基丙二酸(^(18)F-ML-10)PET/CT显像,判定其用于细胞凋亡评估的可能性。材料与方法取荷瘤兔VX2肿瘤细胞动物模型12只,按完全随机化法分为化疗组和对照组,每组6只,化疗组经耳缘静脉输注紫杉醇注射液(按兔体表面积175 mg/m^2计算)诱导肿瘤细胞凋亡,对照组静脉滴注等量生理盐水。2 d后行^(18)F-ML-10 PET/CT全身断层融合显像;同时,对瘤体组织行细胞凋亡百分比检测。分析对照组与化疗组之间瘤体最大标准化摄取值(SUVmax)及瘤体细胞凋亡百分率的差异;分析化疗组瘤体组织SUVmax与肿瘤细胞凋亡百分率之间的相关性。结果化疗组瘤体部位的SUVmax高于对照组(1.65±0.33比0.59±0.17,P=0.004);化疗组细胞凋亡百分率高于对照组[(26.22±5.41)%比(4.55±2.23)%,P=0.004];Spearman秩相关分析显示,化疗组瘤体组织SUVmax值与细胞凋亡百分率呈正相关(r_s=0.943,P=0.005)。结论 ^(18)F-ML-10 PET/CT断层融合显像可用于肿瘤细胞凋亡的评估。Purpose To evaluate the possibility of 18F-ML-10 PET/CT imaging in apoptosis assessment using VX2 tumor cells from tumor-bearing rabbits induced by paclitaxel. Materials and Methods VX2 tumor cells from tumor-bearing rabbits in twelve animal models were collected and were randomly divided into chemotherapy group and control group, with 6 cases in each group. Paclitaxel injection was administered via auricular veins (counted in line with 175 mg/m2 rabbit body surface area) for the chemotherapy group to induce tumor cells apoptosis and control group was injected with the same amount of normal saline via intravenous dripping. 18F-ML-10 PET/CT whole body tomography was performed two days later, and meanwhile apoptotic cell percentage of tumor body tissues was detected. The difference between SUVmax and apoptotic cell percentage of tumor body in control group and chemotherapy group was analyzed. The correlation between SUVmax value and apoptotic cell percentage of tumor body tissues in chemotherapy group was also analyzed. Results SUVmax value of tumor body sites in chemotherapy group was higher than that in control group (1.65±0.33 vs. 0.59±0.17, P=0.004). Apoptotic cell percentage in chemotherapy group was higher than that in control group [(26.22±5.41)% vs. (4.55±2.23)%, P=0.004]. Spearman rank correlation analysis indicated that SUVmax value and apoptotic cell percentage of tumor body tissues in chemotherapy group was in positive correlation (rs=0.943, P=0.005). Conclusion 18F-ML-10 PET/CT tomography can be used for tumor cell apoptosis assessment.
