摘  要:目的:检测卵巢癌患者新型肿瘤标志物LyGDI水平,分析其与肿瘤恶性分子基因表达量的关系。方法:选取2015年1月至2017年5月在南方医科大学附属小榄医院接受手术治疗的卵巢病变患者175例,根据病理结果将其分为卵巢癌组91例,卵巢良性病变组84例。对比两组术前LyGDI水平以及相应病灶组织中增殖基因[Yes相关蛋白1(YAP1)、T-box转录因子2(TBX2)、含KH RNA结合结构域信号转导相关分子3(KHDRBS3)、氧化还原酶的WW结构域(WWOX)]、侵袭基因[胶原三股螺旋重复蛋白1(CTHRC1)、盘状结构域受体1(DDR1)、E74-like factor 5(ELF5)、转移相关基因3(MTA3)、整合素A2(ITGA2)]、自噬基因[Beclin1、微管相关蛋白1轻链3-Ⅱ(LC3-Ⅱ)]表达量的差异,采用Pearson相关分析法分析卵巢癌患者血清LyGDI水平与肿瘤恶性分子基因表达量的关系。结果:卵巢癌组患者血清LyGDI水平及YAP1、TBX2、KHDRBS3、CTHRC1、DDR1、ITGA2mRNA相对表达量高于卵巢良性病变组,而WWOX、ELF5、MTA3、Beclin1和LC3-ⅡmRNA相对表达量低于卵巢良性病变组,差异均有统计学意义(均P<0.05)。卵巢癌患者血清LyGDI水平与以上增殖、侵袭、自噬基因表达量均存在明显相关关系(均P<0.05)。结论:卵巢癌患者血清中LyGDI水平异常增高,且与癌细胞增殖、侵袭及自噬活性相关,或可作为早期评估卵巢癌患者病情的简便指标。Objective: To detect the serum level of LyGDI in ovarian cancer patients and to analyze the correlation between LyGDI and the expression of malignant tumor molecules.Methods: A total of 175 patients with ovarian disease in our hospital from January 2015 to May 2017 were selected,and divided into ovarian cancer group ( n =91) and ovarian benign lesion group ( n =84) according to pathological findings.The preoperative LyGDI content in serum and the expressions of YAP1,TBX2,KHDRBS3,WWOX,CTHRC1,DDR1,ITGA2,ELF5,MTA3,Beclin1 and LC3-Ⅱwere determined and compared.The association between LyGDI level and the mRNA expressions of malignant tumor molecules was analyzed.Results: The serum LyGDI level and YAP1,TBX2,KHDRBS3,CTHRC1,DDR1 and ITGA2 mRNA levels were significantly higher in the ovarian cancer group than those in the ovarian benign lesion group,while the mRNA expression levels of WWOX,ELF5,MTA3,Beclin1 and LC3-Ⅱwere lower ( P 〈0.05).The Pearson correlation analysis showed that serum content of LyGDI in ovarian cancer patients was related to the proliferation,invasion and autophagy related genes expressions. Conclusion: Patients with ovarian cancer had markedly higher level of LyGDI.The serum LyGDI level was correlated with cancer cells’ proliferation,invasion and autophagy activities,and it might be a circulating tumor marker for ovarian cancer.
