富血小板纤维蛋白和浓缩生长因子析出液中相关成骨生长因子含量的比较研究

摘　　要：目的:比较富血小板纤维蛋白(PRF)及浓缩生长因子(CGF)析出液中骨成型蛋白2(BMP-2)、转化生长因子β1(TGF-β1)和碱性成纤维生长因子(bFGF)的释放浓度及规律。方法:无菌条件下分别制备大耳兔PRF膜片和CGF膜片,于细胞培养箱中静置孵育,分别于第0、1、3、5、7天收集PRF和CGF析出液,应用酶联免疫吸附法(ELISA)测定析出液中的BMP-2、TGF-β1和bFGF的浓度并进行分析。结果:PRF组和CGF组析出液中BMP-2、TGF-β1和bFGF的浓度均随着时间推移逐渐增高,于第5天达到峰值后开始降低(P <0. 05);第5天时CGF组析出液中BMP-2、TGF-β1和bFGF的浓度均高于PRF组析出液(P <0. 05)。PRF组和CGF组各时间点析出液中均为BMP-2浓度最高,TGF-β1浓度次之,bFGF浓度最低(P <0. 05)。结论:PRF和CGF析出液中BMP-2、TGF-β1和bFGF 3种生长因子具有相似的释放规律,且同期析出液中生长因子的释放浓度CGF高于PRF。AIM： To investigate the release pattern and concentration of bone morphogenetic protein 2 （BMP-2）, transforming growth factor β1 （TGF-β1） and basic fibroblast growth factor （bFGF） in the extracts of platelet-rich fibrin （PRF） and concentrated growth factor （CGF） precipitates. METHODS： PRF and CGF diaphragm of rabbits were prepared under the aseptic situation and incubated in a cell culture incubator. The PRF and CGF extracts were respectively collected 0, 1, 3, 5 and 7 days after incubation. The concentrations of BMP-2, TGF-β1 and bFGF in the extracts were measured by ELISA, data were statistically analyzed. RESULTS： The concentrations of BMP-2, TGF-β1 and bFGF in the PRF and CGF extracts gradually increased with time passing by and began to decrease after reaching the peak value on the fifth day（ P 〈0.05）. On day 5, the concentrations of TGF-β1 and bFGF in CGF extract were higher than those in PRF extract（ P 〈0.05）. The concentration of BMP-2 was the highest in the extracts of PRF and CGF within 7 days, followed by TGF-β1, and bFGF was the lowest（ P 〈0.05）. CONCLUSION： BMP-2, TGF-β1 and bFGF produced in the extracts of PRF and CGF have a similar release pattern. The concentration of the growth factors in the CGF extract is higher than those of PRF extract in the same period.
