摘 要:为寻找纤维素酶高产菌株实现木质纤维素类生物质的降解及其资源化利用,从常年堆积干枯腐败植物茎秆的林间采集土样进行富集培养2 d后,结合梯度稀释涂平板法和划线分离法进行纤维素酶产生菌的分离纯化.采用刚果红—羧甲基纤维素钠水解圈法作为初筛法、胞外酶活测定法(DNS法)作为复筛法进行菌株的筛选,得到一株产纤维素酶活较高的菌株SL-3.该菌株能够利用羧甲基纤维素钠作为唯一碳源进行生长,酶活达到0. 44 U/m L.通过一系列生理生化实验和16SrDNA序列分析,鉴定该菌株属于肠杆菌属Enterobacter sp.,命名为Enterobacter sp. SL-3.与其他细菌纤维素酶比较发现,该菌株所产纤维素酶活性较高;扫描电镜分析发现,利用该菌株对木质纤维素类生物质进行降解可促进秸秆资源化利用.From the viewpoint of environmental protection and waste recycling, it is important to seek high-yield cellulase-producing bacteria for degrading of lignocellulosic biomass. During the process of screening, the methods of congo red staining and DNS were used. A strain which can use sodium carboxymethyl cellulose (CMC-Na) as sole carbon source was isolated from samples rich in lignocellulosic biomass, and the cellulase activity of 0.44 U/mL was observed. The bacterium isolated was identified as Enterobacter sp. strain by cell morphology,physiology and biochemistry and 16S rDNA sequence analysis appraisal and was named as Enterobacter sp. SL-3. Compared with other bacteria, cellulase activity from SL-3 was relatively higher. Meanwhile the scanning electron microscope (SEM) images demonstrated that the strain Enterobacter sp. SL-3 could accelerate the degradation and realize the resource utilization of lignocellulosic biomass.
