摘 要:目的:探讨γ-生育三烯酚(γ-tocotrienol,γ-T3)在人胃腺癌SGC-7901细胞能量代谢、细胞增殖和凋亡过程中的作用及其机制。方法:用不同浓度(0、15、30、45和60μmol/L)的γ-T3作用SGC-7901细胞后,采用MTT法检测细胞增殖能力的变化。用30μmol/L的γ-T3作用SGC-7901细胞后,采用FCM法检测细胞凋亡情况,采用JC-1荧光探针法检测细胞中线粒体膜电位情况,采用DCFH-DA荧光探针法检测细胞中活性氧(reactive oxygen species,ROS)水平,应用乳酸检测试剂盒检测细胞乳酸含量;采用萤火虫荧光素酶法检测细胞中ATP含量。结果:15~60μmol/Lγ-T3作用SGC-7901细胞24 h后,细胞增殖能力受到明显抑制(P值均<0.05)。用30μmol/L的γ-T3处理后,SGC-7901细胞发生凋亡(P <0.001),且细胞线粒体膜电位明显降低(P <0.001),而细胞中ROS水平明显升高(P <0.001),另外乳酸含量也呈升高趋势(P <0.001),但ATP含量呈降低趋势(P <0.001)。结论:γ-T3可能通过调节人胃腺癌SGC-7901细胞能量代谢来抑制细胞增殖,促进细胞凋亡。Objective: To investigate the effects of γ-tocotrienol (γ-T3) on energy metabolism, proliferation and apoptosis of human gastric adenocarcinoma SGC-7901 cells and their mechanism.Methods: The different concentrations of γ-T3 (0, 15, 30, 45 and 60 μmol/L) were used to treat SGC-7901 cells, then the cell proliferation was detected by MTT assays. After SGC-7901 cells were treated with 30 μmol/L γ-T3, the apoptosis rate was detected by flow cytometry,the mitochondrial membrane potential was detected by JC-1 fluorescence probe, the reactive oxygen species (ROS) level was detected by DCFH-DA fluorescence probe, the lactate level was detected by Lactate Assay Kit, and the content of ATP was quantified by Firefly luciferase. Results: The proliferation of SGC-7901 cells was significantly inhibited by 15-60μmol/L γ-T3 (all P 〈 0.05). After treatment with 30 μmol/L γ-T3, the apoptotic rate of SGC-7901 cells was significantly increased (P 〈 0.001), the mitochondrial membrane potential was significantly decreased (P 〈 0.001), the ROS level was significantly increased (P 〈 0.001), the lactate level was significantly increased (P 〈 0.001), while the content of ATP was significantly decreased (P 〈 0.001). Conclusion: γ-T3 can inhibit the proliferation of human gastric adenocarcinoma SGC-7901 cells, and induce apoptosis through regulating the cell energy metabolism.
