摘 要:目的研究结直肠癌MICA基因的表达与p53、K-ras基因突变之间的联系。方法选取我院从2015年8月~2017年8月132例结直肠癌患者为此次研究对象行回顾性分析,应用半定量PCR-SSCP技术,进行mRNA水平的检测,对结直肠癌组织MICA基因表达量与p53、K-ras进行检测,并分析结直肠癌MICA基因表达与p53、K-ras基因突变之间的相互关联。结果在K-ras基因突变组中的MICA平均表达量显著高于K-ras基因未突变组,差异具有统计学意义(P<0.05);在p53基因突变组中的MICA平均表达量显著高于p53基因未突变组,差异具有统计学意义(P<0.05)。结论高表达的MICA基因的发生可能与p53、K-ras基因突变关系密切,MICA基因的表达情况是目前检测结直肠癌的关键,应引起人们的广泛关注。Objective To study the relationship between the expression of MICA gene in colorectal cancer and the mutations of p53 and K-ras genes. Methods 132 patients with colorectal cancer from August 2015 to August 2017 in ourhospital were selected as the study subjects, and they were retrospectively analyzed. Semi-quantitative PCR-SSCPtechnique was used for the detection at the mRNA level. The expression quantity of MICA gene in colorectal cancertissues and p53 and K-ras were detected. The correlation between MICA gene expression in colorectal cancer and mutations in p53 and K-ras genes was analyzed. Results The average expression quantity of MICA in K-ras gene mutantsgroup was significantly higher than that in K-ras gene non-mutants group, and the difference was statistically significant(P〈0.05); the average expression quantity of MICA in p53 gene mutants group was significantly higher than thatin p53 gene non-mutants group, and the difference was statistically significant(P〈0.05). Conclusion The occurrence ofhighly expressed MICA gene may be closely related to the mutation of p53 and K-ras genes. The expression of MICAgene is the key to the current detection of colorectal cancer and should be widely concerned.
