摘  要:目的制备京尼平交联的^(131)I标记壳聚糖/明胶-拓扑替康膜,并研究复合膜对SK-OV-3荷瘤裸鼠的治疗效果,为同步放化疗载药膜剂的肿瘤治疗等相关研究提供基础。方法制备SK-OV-3腹腔荷瘤裸鼠模型,并手术植入复合膜到荷瘤裸鼠腹腔中,观察各组裸鼠的生存周期、转移瘤及血性腹水重量,考察复合膜对腹腔荷瘤裸鼠的治疗效果。结果 SK-OV-3腹腔荷瘤裸鼠经手术植入复合膜后观察各组生存天数、腹腔转移瘤及血性腹水量可知,同时应用拓扑替康及^(131)I的^(131)I+TPT组疗效优于单独使用拓扑替康或^(131)I,稍次于顺铂对照组,明显优于空白对照组。结论 ^(131)I标记的壳聚糖/明胶拓扑替康载药膜对SK-OV-3腹腔荷瘤裸鼠有一定疗效,能延长荷瘤裸鼠的生存天数并抑制腹腔转移瘤及血性腹水的形成。Objective To prepare Genipin cross-linked 131I-labeled chitosan/gelatin-topotecan membrane, and to investigate the therapeutic effect of the composite membrane on the SK-OV-3 tumor-bearing nude mice, so as to provide abasis for the related research on tumor treatment of concurrent chemoradiotherapy-loaded drug-coated drugs. MethodsSK-OV-3 peritoneal tumor-bearing nude mice model was prepared and surgically implanted into the abdominal cavityof tumor-bearing nude mice. The life cycle, metastatic tumor and blood ascites weight of nude mice in each group wereobserved. The therapeutic effect of composite membrane on abdominal cavity tumor-bearing nude mice was investigated. Results The survival days, abdominal metastases and blood ascites were observed in SK-OV-3 peritoneal tumor-bearing nude mice after surgical implantation of composite membrane. At the same time, the efficacy of the topotecanand 131I in the 131I+TPT group was better than that of topotecan or131I alone, which was slightly lower than that of the cisplatin in the control group, but was significantly better than that in the blank control group. Conclusion 131I-labeled chitosan/gelatin topotecan drug-loaded membrane has a certain curative effect on SK-OV-3 peritoneal tumor-bearingnude mice, which can prolong the survival days of tumor-bearing nude mice and inhibit the formation of abdominal metastases and bloody ascites.
