摘 要:目的:检测采用普通榨汁法和破壁法处理得到的葡萄汁中的化学成分,并研究2种葡萄汁对动物衰老模型的作用。方法:应用HPLC-MS测定2种方法获得的葡萄汁中化合物的含量。给予昆明小鼠皮下注射150mg/kg的D-gal溶液,并饲予单蒸水和120℃烘焙12h后的饲料,制备营养不良的衰老动物模型。小鼠造模第14天随机分为3组,然后每天禁水2h后,自由饮全成分葡萄汁、普通葡萄汁或饮用水6h,其他时间常规饲养,连续7w。实验过程中监测小鼠体重和饮食饮水量。实验终点拍照记录小鼠外观状态,测定小鼠自主活动、跳台潜伏期及错误次数、脏器指数、氧化酶和抗氧化酶含量,并对肾脏进行HE染色,观察病理状态,Western blot检测衰老相关蛋白表达。结果:在相同饮用量的情况下,饮用葡萄汁能够增加模型组小鼠自主活动次数,延长跳台潜伏期、减少错误次数、提高学习记忆能力。其中,饮用全成分葡萄汁能够显著改善模型小鼠增加的肝脏指数,改善肾小球肿胀及肾小管上皮细胞水肿的病理状态,提高抗氧化酶SOD、GSH-Px的含量,抑制脂质过氧化物MDA含量的升高,降低促衰老蛋白FOXO3a、p21的表达,促进抗衰老蛋白Sirt1、Sirt6的表达。结论:葡萄汁可有效抵抗D-gal诱导的小鼠衰老反应,全成分葡萄汁效果要优于普通葡萄汁。ObjectiveTo compare the chemical components release and anti-aging effect of grape treated by common cooking machine and wall broken machine,and study the anti-aging effect.Method The content of two kinds of compounds in grape processing method by HPLC-MS was determined.The D-gal solution was given to Kunming mice by hypodermic injection of 150mg/kg,and given distilled water and baking at 120℃ after 12 hours feed.During the third weeks of modeling,grape juice was treated by different machine in the treatment group.After water-deprivation for 2 hours followed with 6 hours free drinking of grape juice for seven consecutive weeks.The body weight of mice and other parameters were tested during the experiments and at the end of experiments.At the end of experiment,take pictures of the mouse and record the appearance of the mouse.The number of locomotor activities and the lantency of step through and step down as well as the number of errors were tested and recorded.The organ index and the content of the oxidase and antioxidant enzyme were determined.HE staining method was used on the renal of the mouse to observe the pathologic state.Western blot was used to detect the expression of senescence-associate protein.Result At the same amount of drinking,grape juice drinking group could increase the number of locomotor activities of mice caused by D-gal,prolong the latency and reduce mistake times,improve the learning and memory ability of mice.The broken grape juice group could significantly reduce the liver index of mice induced by D-gal,improve the contents of antioxidant enzymes SOD and GSH-Px,inhibit the increase of MDA content in lipid peroxides,improve glomerular swelling and edema of renal tubular epithelial cells induced by D-gal.At the same time,broken grape juice could reduce the expression of senescence promoting proteins FOXO3a and p21,and promote the expression of anti-aging proteins Sirt1 and Sirt6.Conclusion The effect of grape juice with different wall breaking patterns was different,and the grape juice t
