摘　　要：目的 :探讨lncRNA PCGEM1对结肠癌SW480细胞的增殖与侵袭的影响。方法 :培养人结肠癌SW480细胞,将结肠癌细胞系SW480细胞分成3组进行处理,即空白对照组:不转染慢病毒;阴性对照组:转染无序序列慢病毒载体;试验组:转染沉默lncRNA PCGEM1序列慢病毒载体。荧光定量PCR(RT-PCR)检测RNA表达,CCK-8试剂盒、Transwell和AV-PI试剂盒检测增殖、侵袭与凋亡。结果:lncRNA PCGEM1沉默表达慢病毒载体的滴度为1×10~8TU/mL。RT-PCR结果显示:试验组与阴性对照组相比,lncRNA PCGEM mRNA表达较低,差异有统计学意义(q=8.141,P<0.05);Caspase-3 mRNA和Caspase-9 mRNA的表达具有同样的表达趋势(P<0.05)。Transwell实验结果显示,3组之间差异有统计学意义(F=21.982,P<0.05),试验组和阴性对照组比较差异有统计学意义(q=4.223,P<0.05)。CCK-8检测结果显示,3组差异有统计学意义(F=21.214,P<0.05),试验组与阴性对照组比较差异有统计学意义(q=5.272,P<0.05)。AV-PI检测结果显示,3组比较差异有统计学意义(F=31.009,P<0.05),试验组与空白载体病毒组比较差异有统计学意义(q=9.131,P<0.05)。结论:lncRNA PCGEM1在结肠癌细胞中高表达。沉默lncRNA PCGEM1表达可以抑制结肠癌细胞增殖和侵袭,促进凋亡。Objective： To explore the effect of lncRNA PCGEM1 on proliferation and invasion of colon cancer SW480 cells. Methods： Human colon cancer SW480 cells were cultured and divid-ed into three groups according to the treatment methods： blank control group（colon cancer cell line SW480 cell group without lentivirus transfection）, negative control group（colon cancer cell line SW480 cell transfection disordered sequence lentivirus vector） and experimental group（colon cancer cell line SW480 cell transfection disordered sequence lentivirus vector）. SW480 cells were transfected to silence lncRNA PCGEM1 sequence lentiviral vector. RT-PCR was used to detect RNA expression, CCK-8 kit, Transwell and AV-PI kit were used to detect proliferation, invasion and apoptosis. Results： the titer of lentiviral vector expressing lncRNA PCGEM1 was 1 α 108 TU/mL. The results of RT-PCR showed that there was significant difference between the test group and the negative control group（q=8.141, P〈0.05）. The expression of Caspase-3 and Caspase-9 had the same trend. Transwell test results showed that the difference between the 3 groups was statistically significant（F=21.982, P〈0.05）. The difference between the experimental group and the negative control group was statistically significant（q=4.223, P〈0.05）. CCK-8 test showed that the difference between the 3 groups was statistically significant（F=21.214, P〈0.05）. The difference between the experimental group and the negative control group was statistically significant（q=5.272, P〈0.05）. AV-PI test results showed that there were significant differences among the three groups（F=31.009, P〈0.05）, and there were significant differences between the experimental group and the blank vector virus group（q=9.131, P〈0.05）. Conclusion： lncRNA PCGEM1 is highly expressed in colon cancer cells. Silencing lncRNA PCGEM1 expression can inhibit proliferation and invasion of colon cancer cells and promote apoptosis.
